Effect of Jianpi Xiaoai Prescription and Its Decomposed Recipes on Body Weight, Tumors, Metastasis Rate and Expressions of Caspase-3, Bcl-xL, Bax in Colorectal Cancer Model Nude Mice
目的:观察健脾消癌方及其拆方对肠癌模型裸鼠体质量、瘤体、转移率及半胱氨酸天冬氨酸蛋白酶-3(Caspase-3),抗凋亡蛋白B细胞淋巴瘤/白血病-xL (B-cell lymphoma/leukemia-xL,Bcl-xL)和Bcl-2相关X蛋白(Bcl-2 associated X protein,Bax)表达的影响。方法:建立裸鼠肠癌原位瘤模型,分为模型组,健脾益气组(15 g · kg-1),化瘀解毒组(15 g · kg-1),健脾消癌方组(全方组,15 g · kg-1),5-氟尿嘧啶组(5-Fu,20 mg · kg-1),共5组。分别予以相应药物干预4周,观察裸鼠体重、瘤体抑制率、转移率,蛋白免疫印迹法(Western blot)检测Caspase-3,Bcl-xL,Bax蛋白表达。结果:健脾益气组、全方组体重下降低于模型组(P<0.05),化瘀解毒组体重下降高于模型组(P<0.05);健脾益气组对瘤体的抑制与模型组比较无统计学差异,与空白组比较,化瘀解毒组、全方组对瘤体均有较好抑制作用(P<0.05);各组均出现不同程度地转移,其中模型组、健脾益气组总体转移率达90%,化瘀解毒组、全方组转移率分别为70%,60%,低于模型组(P<0.05);与模型组比较,化瘀解毒组及全方组能够上调Bax蛋白表达,下调Caspase-3,Bcl-xL蛋白表达(P<0.01)。结论:健脾消癌方中健脾益气组优势在于稳定体重,化瘀解毒组优势在于抑制肿瘤,抑制转移;全方组能够稳定体质量,抑制肿瘤,抑制转移,全方配伍疗效最优,其抗肿瘤机制可能是上调凋亡相关蛋白Bax及下调Caspase-3,Bcl-xL,诱导肿瘤细胞凋亡。

Objective: To observe the effect of Jianpi Xiaoai prescription and its decomposed recipes on the body weight

metastasis rate and expressions of Caspase-3
B-cell lymphoma/leukemia-xL (Bcl-xL) and Bcl-2 associated X protein (Bax) in colorectal cancer model nude mice.Method: The colorectal cancer model was established
and divided into control group
the Jianpi Yiqi group (15 g · kg-1)
the Huayu Jiedu group (15 g · kg-1)
the Jianpi Xiaoai prescription group (omni-recipe group
15 g · kg-1) and the 5-fluorouracil group (5-Fu
20 mg · kg-1). The body weight
tumor inhibition rate and metastasis rate were observed
and the protein expressions of Caspase-3
Bcl-xL
and Bax of nude mice were detected by Western blot after the corresponding drug intervention for 4 weeks.Result: The weight loss of the Jianpi Yiqi group and the omni-recipe group was less than that of control group (P<0.05)
Huayu Jiedu group' weight loss was higher than that of control group (P<0.05). The Jianpi Yiqi group had no statistical differences from control group in tumor inhibition rate; the Huayu Jiedu group and the omni-recipe group had a better tumor inhibition rate (P<0.05). All of the groups had different degrees of metastasis. Specifically
the control group and the Jianpi Yiqi group had the overall metastasis rate of 90%
and the Huayu Jiedu group and the omni-recipe group had the metastasis rate of 70% and 60%
respectively
both of which were lower than that of control group (P<0.05). Compared with control group
the Huayu Jiedu group and the omni-recipe group showed increase in the protein expression of Bax
and decrease in the protein expressions of Caspase-3
Bcl-xL (P<0.01).Conclusion: The Jianpi Yiqi group shows a stable weight
while the Huayu Jiedu group shows inhibitions in tumor and metastasis. The omni-recipe group shows a stable weight and inhibitions in tumor and metastasis
with the best effect. The antitumor mechanism of Jianpi Xiaoai prescription may be correlated with the up-regulation in expressions of apoptosis-associated proteins Bax and the down-regulation in Caspase-3
