Protective Effect of Shenfu Injection on Myocardium of Rats with Severe Acute Pancreatitis
目的:探讨参附注射液治疗重症急性胰腺炎模型大鼠心肌的保护作用。方法:选取雄性SD大鼠60只,随机分为6组,分别为假手术组、模型组、乌司他丁组(2×104 U · kg-1)和参附注射液高、中、低剂量组(3,1.5,0.75 g · kg-1)。实验采用逆行胆胰管灌注5%牛磺胆酸钠溶液复制重症急性胰腺炎大鼠模型,造模后3 h,分别给予尾静脉注射生理盐水、乌司他丁、不同剂量参附注射液。6 h后苏木素-伊红(HE)染色法观察各组大鼠胰腺及心肌的病理变化,并检测大鼠心肌细胞Na+-K+-ATP酶活性,实时荧光定量聚合酶链式反应(qPCR)法检测肿瘤坏死因子-α(TNF-α),白细胞介素-1β(IL-1β)和白细胞介素-6(IL-6)mRNA表达量。结果:与假手术组比较,模型组的大鼠胰腺、心肌损伤严重,病理评分明显升高(P<0.05),心肌Na+-K+-ATP酶活性明显降低(P<0.05),TNF-α,IL-1β mRNA表达量明显升高(P<0.05);与模型组比较,乌司他丁组、参附注射液组的胰腺和心脏病理评分明显降低(P<0.05),参附注射液组大鼠心肌Na+-K+-ATP酶活性明显增加(P<0.05),TNF-α,IL-1β mRNA表达量明显降低(P<0.05)。结论:参附注射液可能通过改善心肌细胞Na+-K+-ATP酶活性,减少心肌组织中TNF-α,IL-1β的表达,对心肌起保护作用。
Abstract
Objective: To investigate the protective effect and mechanism of Shenfu injection on myocardium in rats with severe acute pancreatitis. Method: Totally 60 male SD rats were randomly divided into 6 groups:Sham operation group
model group
ulinastatin (2×104 U · kg-1) group and Shenfu injection groups (3

0.75 g · kg-1). The models of severe acute pancreatitis were established by retrograde cholangiopancreatic injection of 5% sodium taurocholate solution. Three hours after modeling
the normal saline
ulinastatin and Shenfu injection were respectively given by tail vein injection. After 6 hours
the pathological changes of pancreas and myocardium were observed; the expression of Na+-K+-ATPase activity was detected; and the mRNA expression levels of tumor necrosis factor-α (TNF-α)
interleukin-1β (IL-1β) and interleukin-6 (IL-6) in rat cardiomyocytes were detected by Real-time fluorescent quantitative polymerase chain reaction (qPCR). Result: As compared with the sham operation group
the pancreatic and myocardial injuries were severe and pathological score was significantly increased in model group (P<0.05); the activity of Na+-K+-ATPase was significantly decreased (P<0.05)
and the mRNA expression levels of TNF-α and IL-1β were significantly increased (P<0.05). As compared with the model group
the pancreatic and cardiac pathological scores were significantly decreased in Ulinastatin group and Shenfu injection groups (P<0.05); the activity of Na+-K+-ATPase in the myocardium was significantly increased and the mRNA expression levels of TNF-α and IL-1β were significantly decreased in Shenfu injection group (P<0.05). Conclusion: Shenfu injection may improve the activity of Na+-K+-ATPase in myocardial cells and decrease the mRNA expression of TNF-α and IL-1β in myocardium to protect the myocardium.
