Effect of Extract on Expressions of AQP-4 and NGF After Intracerebral Hemorrhage in Rats
目的:研究藏荆芥提取物治疗脑出血模型大鼠脑水肿和脑血肿的作用,并探讨其可能的机制。方法:采用胶原酶注射尾状核法建立脑出血大鼠模型,随机分为正常组、假手术组、模型组、藏荆芥高、中、低剂量(6.4,3.2,1.6 g ·kg-1)组、尼莫地平组。各组大鼠自给药开始计时,7 d后观察大鼠行为,通过Longa评分法对各组大鼠进行神经行为学评分;之后处死大鼠,取大鼠出血侧大脑半球测定湿重,烘干后测定干重,通过多田公式计算大鼠脑含水量;取大鼠脑组织并制作成切片,观察切片测定大鼠的脑血肿体积;并采用免疫荧光法检测大鼠脑部的水通道蛋白-4(aquaporin-4,AQP-4)和神经生长因子(nerve growth factor,NGF)的表达情况。结果:藏荆芥提取物可以明显减轻脑出血大鼠的神经行为学评分(P<0.05,P<0.01),藏荆芥大剂量组可以明显减轻脑水肿(P<0.05,P<0.01),但对血肿的清除并无明显作用。与模型组比较,藏荆芥使大鼠脑部AQP-4的表达显著降低以及NGF的表达明显增加(P<0.05)。结论:藏荆芥提取物能改善脑出血模型大鼠的神经行为,有减轻脑出血模型大鼠脑水肿的作用,藏荆芥通过抑制大鼠脑部AQP-4的表达,促进NGF的表达,从而治疗大鼠的脑出血。
Abstract
Objective: To study the effect of Nepeta angustifolia extract on hematoma in intracerebral hemorrhage model rats and its possible mechanism. Method: The intracerebral hemorrhage model was established by injecting with collagenase. Male SD rats were divided randomly into normal control group

the neurobehavioral score of each group was evaluated by Longa score method. Subsequently
the rats were put to death
and the hemorrhagic hemisphere was measured to determine the wet weight. After drying
the dry weight of the rat brain was measured
and the brain water content of the rat brain was calculated by the Tada formula. The brain tissues of the rats was collected and made into slices. The volume of cerebral hematoma of the rats was observed by sections. Immuno fluorescence was used to observe the expressions of aquaporin-4 (AQP-4) and nerve growth factor (NGF). Result: The neurological behavior score of N. angustifolia groups was lower than that of the model group (P<0.05). The hematoma was not significantly removed
the brain edema of large-dose N. angustifolia group was significantly reduced (P<0.05). Compared with the model group
the expression of AQP-4 in rats treated with N. angustifolia decreased significantly (P<0.05)
the expression of NGF of the medium-dose group increased significantly (P<0.05
P<0.01). Conclusion: N. angustifolia extract has a protective effect on neurobehavioral score and content of brain water
