摘要

目的：探索葛根芩连汤含药血清对胰岛素抵抗（IR）的HepG2（IR-HepG2）细胞糖代谢的调控关系。方法：将正常的HepG2细胞作为空白组，IR-HepG2细胞分为模型组，非诺贝特组，葛根芩连汤低、中、高剂量组，以葡萄糖消耗量为药效指标，采用色谱-质谱联用技术，对IR-HepG2细胞及给药后的IR-HepG2细胞内代谢产物进行分析，经过Mass Profiler Professional（MPP）软件对数据进行分析，得出生物标志物，验证之后，研究呈剂量依赖的生物标志物对糖耗量的关系。结果：发现葛根芩连汤含药血清能提高IR-HepG2细胞的糖耗量，其调节机制可能和文中6个生物标记物有关，已确定其中3个生物标志物是存在剂量依赖性且对糖代谢具有调控作用。结论：葛根芩连汤含药血清可能通过调节IR-HepG2细胞色氨酸，泛酸和腺嘌呤的量达到提高糖代谢的作用。

Abstract

Objective: To explore the regulatory effect of Gegen Qinlian Tang serum on the glucose metabolism of insulin resistance (IR) HepG2 (IR-HepG2). Method: Normal HepG2 cells were taken as blank control group. IR-HepG2 cells were divided into model group

the relationship between glucose consumption and biomarkers was studied. Result: Three biomarkers were found to be dose dependent and could regulate glucose metabolism. Conclusion: Gegen Qinlian Tang-containing serum could regulate the content of tryptophan

pantothenic acid and adenine in IR-HepG2 cell to reduce sugar metabolism.
