Effects of Suoquan Wan on -ARmRNA Expression of Detrusor in Natural Aging Rats
Objective: The mechanism of Suoquan Wan(SQW) reducing urination was investigated by observing the change of the expression of β3- adrenergic receptor(AR)of mRNA in bladder detrusor of the natural aging rat. Method: Fifty aged rats (20 months) and 10 young rats (3 months) were chosen. Young rats were chosen as control
aged rats were randomly divided into five groups: model
Jinkui Shenqi pill group
SQW high
middle and low dose group
10 rats in each group. The low
middle and high dose group of SQW were given the appropriate concentration (0.293
0.585
1.170 g ·kg-1 ·d-1). Jinkui Shenqi pill group was given appropriate liquid(1.08 g ·kg-1 ·d-1)
once everyday for four weeks. The urination after the water load in rats at 5 h and 24 h was measured in metabolic cages.β3-AR mRNA expression was detected by PCR in the detrusor of the natural aging rats. Result : The 24 h urination of SQW middle and high dose group significantly decreased. The 5h urination after the water load in rats of SQW low
middle and high dose group also significantly decreased.β3-AR mRNA expression significantly decreased in the detrusor of the natural aging rats(P<0.05). SQW high dose can remarkably elevate β3-AR mRNA expression in the detrusor of the natural aging rats after administration SQW. Conclusion : SQW can remarkably elevate β3-AR mRNA expression in the detrusor of the natural aging rats
which was the possible mechanism of SQW reducing urination.
