      Abstract:ObjectiveTo investigate the effect of microemulsion on the distribution of index components in different phases of Zexietang extract based on high performance liquid chromatography(HPLC) and phase separation process.MethodParticle size meter and transmission electron microscope were used to characterize the colloidal particles in blank microemulsion, aqueous extract of Zexietang and microemulsion extract of Zexietang. The phase separation process was established by high-speed centrifugation and dialysis, and based on this process, the aqueous extract and microemulsion extract of Zexietang were separated into the true solution phase, the colloidal phase and the precipitation phase, respectively. The contents of six components, including atractylenolide Ⅲ, atractylenolide Ⅱ, 23-acetyl alisol C, alisol A, alisol B and alisol B 23-acetate, were determined by HPLC with the mobile phase of water(A)-acetonitrile(B) for gradient elution(0-5 min, 40%-43%B; 5-20 min, 43%-45%B; 20-45 min. 45%-60%B; 45-75 min, 60%-80%B). The solubility of the index components in water and microemulsion was determined by saturation solubility method.ResultThe colloidal particles in the aqueous extract, microemulsion extract and blank microemulsion were all spherical, and the particle size, polydispersity index(PDI) and Zeta potential of the colloidal particles were in the order of aqueous extract >microemulsion extract >blank microemulsion. The results of phase separation showed that the colloidal phase and the true solution phase could be completely separated by dialysis for 2.5 h, and the phase separation process was tested to be stable and feasible. Compared with the aqueous extract of Zexietang, the use of microemulsion as an extraction solvent could increase the contents of atractylenolide Ⅲ, 23-acetyl alisol C, atractylenolide Ⅱ , alisol A, alisol B and alisol B 23-acetate by 3.75, 6.82, 35.47, 10.66, 35.41, 27.75-fold, and could increase the extraction efficiencies of the latter five constituents by 2.03, 1.15, 1.70, 6.43, 5.53 times. The solubility test showed that the microemulsion could significantly improve the solubility of atractylenolide Ⅱ, alisol A, alisol B and alisol B 23-acetate, but it had less effect on the solubility of atractylenolide Ⅲ and 23-acetyl alisol C.ConclusionMicroemulsion can improve the extraction efficiency and increase the distribution of the index components in the colloidal phase state of Zexietang to different degrees, providing a reference for the feasibility of microemulsion as an extraction solvent for traditional Chinese medicine.

      • Abstract:Under the national policy background of traditional Chinese medicine(TCM)registration review and approval reform, TCM preparations in medical institutions(hereinafter referred to as medical institution preparations) have human use experience, conform to the characteristics of TCM clinical practice, and have advantages in the research and development of innovative Chinese medicines. Therefore, the research and development mode based on clinical experience prescription-medical institution preparations-innovative Chinese medicines has attracted widespread attention from the industry. However, in the process of development and use of medical institution preparations, there are generally clinical problems that restrict their transformation into innovative Chinese medicines, resulting in a relatively weak collection basis of human use experience and insufficient clinical evidence for supporting the research and development of new TCM medicines. In this paper, on the basis of sorting out the supporting regulations and relevant technical requirements of human use experience, and analyzing the clinical problems restricting the transformation of medical institution preparations, it provides suggestions for medical institutions to carry out high-quality research on human use experience of preparations in the process of TCM clinical practice from the aspects of continuously exploring clinical value, improving the construction of information system, focusing on the TCM clinical practice and giving full play to the advantages of discipline cooperation. By realizing the whole life cycle management of medical institution preparations based on three-combination evaluation evidence system, we can promote the transformation of medical institution preparations into innovative Chinese medicines.

          Abstract:ObjectiveTo evaluate the pharmacological effect of Alismatis Rhizoma (AR) and its processed product on rats with edema of kidney Yin deficiency and explore the mechanism.MethodA total of 42 male SPF SD rats were randomized into normal group (equivalent volume of distilled water), model group (equivalent volume of distilled water), positive medicine Liuwei Diguangwan group (1.4 g·kg-1), low- and high-dose AR groups (1, 4 g·kg-1, respectively), and low- and high-dose salt-processed AR (SAR) groups (1, 4 g·kg-1, respectively), with six rats in each group. Adriamycin (tail vein injection) and thyroxine (gavage) were used to induce edema of kidney Yin deficiency in rats except the normal group. The administration lasted 4 weeks for all the groups. After the last administration, histopathological changes of rat kidneys were observed based on hematoxylin-eosin (HE) staining. Serum content of triiodothyronine (T3), thyroxine (T4), follicle-stimulating hormone (FSH), and testosterone (T) was determined by radioimmunoassay, and serum content of creatinine (CREA), urea (UREA),cholesterol (CHOL) and triglyceride (TG) by automatic biochemical analyser. The levels of gonadotropin-releasing hormone (GnRH), cyclic adenosine monophosphate (cAMP), and cyclic guanosine monophosphate (cGMP) in plasma were measured by enzyme-linked immunosorbent assay (ELISA), and the expression of aquaporin(AQP)-1 and AQP-2 and the transcription of mRNA in kidney were measured by immunohistochemistry and real-time fluorescent quantitative polymerase chain reaction (Real-time PCR), respectively.ResultCompared with normal group, the rats in model group showed decrease in body mass and urine volume (P<0.01), increase in water consumption (P<0.05), infiltration of a large number of inflammatory cells and fibrous tissue proliferation in the kidney, rise of the expression and transcript levels of T3, T4, cAMP/cGMP, CREA, FSH, AQP-1, and AQP-2 (P<0.01), the contents of CHOL and TG were significantly increased (P<0.05), and reduction in the levels of GnRH and T (P<0.01). Body mass increased in both the low- and high- dose groups of AR and SAR compared with that in model group, with significant differences between the low-dose AR group and the low-dose SAR group (P<0.01). Moreover, compared with model group, low- and high-dose AR and SAR insignificantly increased the urine volume of rats, reduced the inflammatory cells in kidney tissues, significantly decreased the levels of T4, cAMP/cGMP, UREA, CREA, FSH, CHOL and TG in serum (P<0.05,P<0.01), and elevated the level of GnRH (P<0.01), high-dose AR, low- and high-dose SAR significantly lowered the transcription levels of AQP-1 and AQP-2 mRNA in the kidneys of rats (P<0.01).ConclusionBoth AR and SAR alleviated the edema of kidney Yin deficiency in rats by down-regulating the expression of AQP-1 and AQP-2 and correcting the hypothalamic-pituitary-gonadal (HPG) axis disorder.

            Abstract:Jianwei Xiaoshi tablets, as a common variety of Chinese patent medicine with "one product with many manufacturers", have many manufacturers and huge market sales. However, the phenomenon about uneven quality and discrepant price is prominent. Based on this, this study was carried out for the quality evaluation of Jianwei Xiaoshi tablets by applying the high-quality evaluation criteria with the quality as core for Chinese patent medicine, which was based on the full production cycle, from the multi-dimension including raw material selection, production process, quality control, post-marketing research and so on. The evaluation results showed that the quality evaluation scores of Jianwei Xiaoshi tablets from different manufacturers varied greatly (ranging from 35 to 66), indicating that the quality was significantly different. In the actual production, generally inadequate attention was paid to the quality of raw materials, and the quality of raw materials was insufficient with the score ratio of 43%, especially the poor consistency control of them. The role of good manufacturing practice was obvious, and the scores of production process were generally high with the average score ratio of 62%, and the maximum up to 80%. The technological advancement of the manufacturer was outstanding. The score ratio of quality control was only 31% that the internal quality standard of each manufacture almost stayed at the qualified line, which was equal to the national standard, and the consistency of products was insufficient. The post-marketing research was lacking with the score ratio of 37%. Manufacturers with high brand awareness and market share were upper scores, while the others lagged far behind. The results of this evaluation are in line with the overall prediction, which can provide a reference for the high-quality evaluation of Chinese patent medicine, and supply the scientific data for high-quality and high-price application.

              Abstract:ObjectiveTo observe the effects of Chaihu Shugansan on gastric motility and mitochondrial function and mitophagy in rats with functional dyspepsia (FD) and to preliminarily reveal its mechanism in preventing and treating FD.MethodThirty-two SD rats of SPF grade were randomly divided into the normal group, model group, Chaihu Shugansan (4.8 g·kg-1) group, and domperidone group (4.5 mg·kg-1) after one week of adaptive feeding, with eight rats in each group. Rats in all groups except for the normal group were exposed to modified tail-clamping stimulation for inducing FD. Four weeks later, the semi-solid nutritive medium was used to observe the gastric emptying rate of FD rats, and the serum citrate synthase (CS), motilin (MTL), and gastrin (GAS) were detected by enzyme-linked immunosorbent assay (ELISA). The pathological changes in gastric tissue were observed by hematoxylin-eosin (HE) staining, the mitochondrial characteristics by transmission electron microscopy, and the expression of microtubule-associated protein light chain 3 (LC3) and voltage-dependent anion-selective channel 1 (VDAC1) by immunofluorescence co-localization. The mitochondria were extracted from fresh gastric tissue for measuring the reactive oxygen species (ROS), malondialdehyde (MDA), and superoxide dismutase (SOD) levels using the biochemical kits. The mitochondrial LC3, yeast Atg6 homologous(Beclin1), and p62 protein expression was assayed by Western blot.ResultCompared with the normal group, the model group exhibited significantly reduced gastric emptying rate (P<0.01) and serum CS, MTL, and GAS levels (P<0.01). HE staining showed no pathological changes like erosion or ulcer in the gastric tissue of rats in each group. However, the mitochondria in the gastric tissue were observed swollen and dilated under the transmission electron microscope, and vacuolar lesions appeared. The co-expression of LC3 with VDAC1 increased significantly (P<0.01). The mitochondrial ROS and MDA levels were elevated significantly (P<0.01), while SOD content was significantly reduced (P<0.01). The protein expression levels of LC3 and Beclin1 were significantly up-regulated (P<0.05, P<0.01), whereas p62 protein expression was down-regulated significantly (P<0.01). Compared with the model group, Chaihu Shugansan and domperidone elevated the gastric emptying rate (P<0.05) and serum CS, MTL, and GAS levels (P<0.05, P<0.01). It was observed under the transmission electron microscope that the mitochondrial nuclear membrane of the gastric tissue was intact, with clear structure and high density of mitochondrial cristae, and mitochondrial fission and fusion were observed in some parts. The co-localization area of LC3 and VDAC1 was significantly reduced (P<0.01). The mitochondrial ROS and MDA levels declined significantly (P<0.05, P<0.01), whereas SOD level rose (P<0.05). The protein expression levels of LC3 and Beclin1 were significantly down-regulated (P<0.05, P<0.01), while that of p62 was obviously up-regulated (P<0.01).ConclusionThe mechanism of Chaihu Shugansan in preventing and treating FD may be related to the improvement of mitochondrial function and the inhibition of mitophagy in gastric tissue.

                Abstract:ObjectiveTo explore the effect of Chaihu Jia Longgu Muli Tang on the hippocampus of rats with chronic stress depression based on the brain-derived neurotrophic factor (BDNF)/tyrosine kinase B (TrkB)/cyclic adenosine phosphate response element-binding protein (CREB) pathway.MethodSixty SD rats were divided into a blank group (n=10) and an experimental group (n=50) for the induction of the chronic stress depression model. The rats in the experimental group were further divided into the following five groups: a model group, a fluoxetine hydrochloride group (0.003 g·kg-1), and low-(1.625 g·kg-1), medium-(3.25 g·kg-1), and high-dose (6.5 g·kg-1) Chaihu Jia Longgu Muli Tang groups. The rats were administered correspondingly by gavage once a day for eight weeks. Behavioral tests were performed to evaluate the depression state of the rats before modeling, after modeling, and after drug administration. Hematoxylin-eosin (HE) staining was used to observe the morphological changes in the hippocampus of rats. The immunohistochemical (IHC) staining was used to quantitatively detect BDNF protein expression in the rat hippocampus. The mRNA and protein expression of BDNF, TrkB, and CREB in the rat hippocampus was detected by the real-time fluorescence-based quantitative PCR (Real-time PCR) and Western blot, respectively.ResultCompared with the blank group, the model group showed decreased sucrose preference rate (P<0.05), declining horizontal and vertical scores (P<0.05), and prolonged immobility time and floating time (P<0.05). Additionally, HE staining results revealed that hippocampal neuron structure was damaged. IHC staining showed that the mRNA and protein expression of BDNF, TrkB, and CREB was significantly decreased (P<0.05). Compared with the model group, the fluoxetine hydrochloride group and the Chaihu Jia Longgu Muli Tang groups displayed elevated sucrose preference rate (P<0.05), increased horizontal and vertical scores (P<0.05), and shortened immobility time and floating time (P<0.05). Furthermore, the hippocampal neuron structure was significantly repaired. IHC staining showed that the mRNA and protein expression of BDNF, TrkB, and CREB was significantly increased (P<0.05).ConclusionChaihu Jia Longgu Muli Tang can significantly improve the depression-like behaviors of rats after chronic stress stimulation and enhance the regeneration and repair of neurons in the hippocampus. The underlying mechanism may be related to the up-regulation of the BDNF/TrkB/CREB signaling pathway in the hippocampus of rats.

                  Abstract:ObjectiveTo establish the high performance liquid chromatography (HPLC) fingerprint of Citri Sarcodactylis Fructus, and to search for makers to characterize the quality difference of Citri Sarcodactylis Fructus from different origins coupled with chemometrics.MethodThe analysis was performed on a Thermo Hypersil GOLD C18 column (4.6 mm×250 mm, 5 μm) with mobile phase consisted of acetonitrile-0.05% phosphoric acid solution for gradient elution, and the detection wavelength was set at 254 nm. A total of 31 batches of samples were analyzed to establish the HPLC fingerprint of Citri Sarcodactylis Fructus. Similarity evaluation was performed by Traditional Chinese Medicine Chromatographic Fingerprint Similarity Evaluation System (2012 edition) to confirm the common peaks, which were identified by comparison of reference substances. On the basis, chemometrics methods were used to analyze and evaluate the quality of Citri Sarcodactylis Fructus from different origins. At the same time, 3 batches of 5 species of decoction pieces from the genus Citrus in the family Rutaceae, including Citri Sarcodactylis Fructus, Aurantii Fructus Immaturus, Aurantii Fructus, Citri Reticulatae Pericarpium Viride and Citri Reticulatae Pericarpium, were randomly collected for evaluating the effectiveness and reliability of the established HPLC fingerprint of Citri Sarcodactylis Fructus.ResultHPLC fingerprint of Citri Sarcodactylis Fructus was established and 22 common peaks were identified. And seven common peaks among them were identified as 6,7-dimethoxycoumarin, diosmin, hesperidin, byakangelicin, 5,7-dimethoxycoumarin, bergapten and oxypeucedanin. Except for 2 batches of samples, the similarities of fingerprints between other 29 batches of samples were >0.9. The 31 batches of Citri Sarcodactylis Fructus were basically divided into 3 groups by cluster analysis and principal component analysis, which were consistent with the classification of three different producing areas. Eight differential markers were screened by orthogonal partial least squares discriminant analysis and four of them (5,7-dimethoxycoumarin, bergapten, 6,7-dimethoxycoumarin and diosmin) were identified by reference substances. Similarity evaluation of 5 species of decoction pieces from genus Citrus in the family Rutaceae was carried out by taking the reference fingerprint of Citri Sarcodactylis Fructus as treference chromatogram, similarity of Citri Sarcodactylis Fructus decoction pieces was 0.892-0.977, and the similarities of the other 4 kinds of decoction pieces were 0.215-0.517.ConclusionThe established fingerprint method is reasonable, effective and accurate for quality control of Citri Sarcodactylis Fructus, the characterization information is more comprehensive combined with chemometrics.

                    Abstract:ObjectiveTo investigate the anti-anxious depression mechanism of Baihe Dihuangtang from the NOD-like receptor thermal protein domain 3 (NLRP3) inflammasome.MethodFifty SD rats were randomly divided into normal group, model group, venlafaxine group (13.5 mg·kg-1), Baihe Dihuangtang high and low dose group (16,4 g·kg-1), with 10 rats in each group. Chronic restraint stress for 28 days (6 h) combined with subcutaneous injection of corticosterone (30 mg·kg-1) was used to establish induce an anxious depression model. From the 8th day of modeling, the rats in the normal group and the model group received distilled water, and those in groups with drug intervention were treated with corresponding drugs by gavage for 21 days. Elevated plus maze and open field test were used to evaluate the behavioral changes of rats. Enzyme- linked immunosorbent assay (ELISA) was used to detect serum and hippocampal interleukin-1β (IL-1β), interleukin-6 (IL-6) and interleukin-18 (IL-18) levels. Western blot were used to detect the relative expression of hippocampal NLRP3, apoptosis-associated speck-like protein containing a CARD (ASC), and Caspase-1. The pathological changes of the hippocampus were observed by hematoxylin-eosin(HE) staining, the average fluorescence intensity of NLRP3, ASC, and Caspase-1 was detected by immunofluorescence. The ultrastructure of neurons was observed under electron microscopy.ResultCompared with the normal group, the model group showed reduced total entries (TE), the ratio of open-arm entries (OE%), the ratio of open-arm times (OT%), and the autonomous activity score (P<0.01), significant anxiety and depression-like behaviors, increased levels of IL-1β, IL-6, and IL-18 in the serum and hippocampus (P<0.01), elevated protein expression of NLRP3, ASC, and Caspase-1 (P<0.01), activated NLRP3 inflammasomes, and injured hippocampal neurons. Compared with the model group, the high-dose Baihe Dihuangtang group showed improved anxiety and depression-like behaviors (P<0.01), and decreased levels of IL-1β, IL-6, and IL-18 in the serum and hippocampus (P<0.05,P<0.01), reduced protein expression of NLRP3, ASC, and Caspase-1 (P<0.01), and alleviated hippocampal neuron damage.ConclusionBaihe Dihuangtang can improve neuronal damage in anxious depression by inhibiting the excessive activation of NLRP3 inflammasomes.

                      Abstract:Polygonati Rhizoma was first recorded in Mingyi Bielu(《名医别录》) as both medicine and food due to its high edible and medicinal values.However,there is no systematic textual research on its use as medicine and food. Polygonati Rhizoma and Polygonati Odorati Rhizoma show similar morphology,so they are often confused.Clematis apiifolia and Polygonatum odoratum are often regarded as the original plants of Polygonati Odorati Rhizoma. By reviewing the related records in Chinese herbal books of the past dynasties,this paper aimed to clarify the relationship between Polygonati Rhizoma,C.apiifolia and P.odoratum,trace the historical evolution of Polygonati Rhizoma as medicine and food from the name,harvesting and processing,property,flavor and meridian tropism,efficacy and indications,edible and medicinal records,and contraindications,and summarize its application in ethnic medicine,in order to inherit and develop the ethnic medicine. The findings demonstrated that the name of Polygonati Rhizoma and its synonyms were derived from its pictogram,function,and place of origin,containing a wealth of myth implication and Taoism thought.Polygonati Rhizoma,C.apiifolia and P.odoratum are three species,different from the conclusions in previous textual research. The seedlings,flowers,seeds,and rhizomes of Polygonati Rhizoma all could be used for edible and medicinal purposes,with its edible records exceeding the medicinal records. There were 22 medical books from the ethnic minority groups such as The Annals of National Medicine in Yunnan,Zhongguo Minzu Yaozhi Yao,and Dictionary of Chinese Ethnic Medicine recording the knowledge and application of 12 species in Polygonatum by 26 ethnic minorities.Due to their different living environments,lifestyles,and cultural customs,ethnic minorities have formed their unique medical cultures.

                        Abstract:Traditional Chinese medicine (TCM) is a great treasure house, exhibiting unique advantages in the treatment of some difficult and critical diseases. The incidence rate of membranous nephropathy has increased year by year in recent years, and has become the first cause of primary glomerular diseases. However, its pathogenesis is not clear. Modern medicine often uses immunosuppressive therapy, but it often faces the problems of high side effects and high recurrence rate. The China Association of Chinese Medicine (CACM) invited clinical experts of TCM and western medicine to fully discuss membranous nephropathy, which was later confirmed to be one of the clinical diseases responding specifically to TCM. Apart from summarizing the pathogenesis and clinical diagnosis and treatment of membranous nephropathy in both TCM and western medicine, this paper also detailed TCM cognition, syndrome differentiation, and therapeutic schemes of membranous nephropathy, aiming to improve the clinical remission rate of membranous nephropathy and provide reference for its clinical treatment.
