- Abstract:ObjectiveTo investigate the therapeutic effect of Yupingfeng San on allergic rhinitis (AR) and its effect on Reactive oxygen species (ROS)/NOD-like receptor thermal protein domain associated protein 3 (NLRP3)/cysteinyl aspartate specific proteinase-1 (Caspase-1) pathway.MethodSPF mice were randomly divided into control group, model group, loratadine group (0.9 mg·kg-1), and low, medium, and high dose Yupingfeng San groups (6, 12, 24 mg·kg-1), with 10 mice in each group. The control group was given routine feeding, and the other groups were intraperitoneally injected with [ovalbumin(OVA) + Al(OH)₃ + phosphate buffer solution(PBS)] suspension once every other day for seven consecutive times. After seven days, 10% OVA solution was instilled in the nose, two times each day for seven consecutive days. After successful modeling, each administration group was intraperitoneally injected with different doses of Yupingfeng San Decoction, and the control group and model group were intraperitoneally injected with an equal volume of normal saline. Symptoms of sneezing, scratching, and runny nose were recorded and scored daily. The levels of ovalbumin specific immunoglobulin E (OVA-sIgE), histamine, eosinophil cationic protein (ECP), prostaglandin D2 (PGD2), interleukin-1β (IL-1β), interleukin-18 (IL-18), interleukin-4 (IL-4), and γ interferon (IFN-γ) in nasal lavage solution and serum of mice were detected by enzyme-linked immunosorbent assay (ELISA). The damage status of the nasal mucosa was observed by hematoxylin-eosin (HE) staining. The number of goblet cells in the nasal mucosa of mice was observed by periodic acid-Schiff (PAS) staining. The expression of NLRP3 protein in the nasal mucosa of mice was detected by immunohistochemistry. Western blot was used to detect the expressions of NLRP3, cleaved Caspase-1, and cleaved gasdermin D (GSDMD) proteins in the nasal mucosa. The test kit was used to detect the changes in ROS in the nasal cavity of mice in each group.ResultCompared with the control group, the nasal symptoms of the model group were significantly aggravated, and the levels of inflammatory factors OVA-sIgE, histamine, ECP, PGD2, IL-1β, IL-18, and IL-4 in serum and nasal lavage solution were increased (P<0.05,P<0.01). The levels of IFN-γ were decreased (P<0.05,P<0.01). The histopathological score, goblet cell number, and ROS content were significantly increased (P<0.01), and the expressions of pyrodeath-related proteins NLRP3, cleaved Caspase-1, and cleaved GSDMD were increased (P<0.01). Compared with the model group, the nasal symptoms of the loratadine group and Yupingfeng San groups were significantly relieved, and the levels of inflammatory factors OVA-sIgE, histamine, ECP, PGD2, IL-1β, IL-18, and IL-4 in serum and nasal lavage solution were decreased (P<0.05,P<0.01). The levels of IFN-γ were increased (P<0.05,P<0.01). The histopathological scores, goblet cell number, and ROS content were significantly decreased (P<0.05,P<0.01), and the expressions of pyrodeath-related proteins NLRP3, cleaved Caspase-1, and cleaved GSDMD were decreased (P<0.05,P<0.01). <HTML><H-PDF><L-PDF>Updated:2023-11-20 - Abstract:ObjectiveTo clarify the intervention effect of Osteoking (OK) in rats with myofascial pain syndrome (MPS) and preliminarily explore the pharmacological mechanism of OK in relieving chronic pain from the perspective of anti-inflammatory disease.MethodThe 60 SD rats were divided into normal group, model group, low, medium, and high dose OK groups (0.66, 1.31, 2.63 mL·kg-1), and positive celecoxib group (21 mg·kg-1). The MPS rat model was established by beating combined with the centrifugal exercise method, and the OK and celecoxib were given at the same time. SMALGO paw pressure pain manometer detected the shock pain point tenderness threshold of rats, and the Von-Frey needle and acetone stimulation method detected the mechanical hyperalgesia threshold and cold hyperalgesia stimulation response respectively. Eight weeks and 10 weeks after modeling, the spontaneous discharge state and convulsion response of MPS rats were determined by electromyograph (EMG) instrument. The gait changes of MPS rats were detected using a CatWalk gait analyzer. The expression levels of interleukin-1 β (IL-1β), tumor necrosis factor-α (TNF-α), substance P (SP), and bradykinin (BK) were measured by enzyme-linked immunosorbent assay (ELISA). The protein expression levels of nuclear transcription factor-κB (NF-κB) inhibiting protein α (IκBα), phosphorylates (p)- IκBα, NF-κB p65, and p-NF-κB p65 were detected in MPS rats by Western blot. The positive expression of p-NF-κB p65 was detected by immunofluorescence.ResultCompared with the normal group, the model group shows 100% positive rates for EMG signal and local convulsions response at both the 8th and 10th weeks. The tenderness threshold and mechanical hyperalgesia threshold are significantly reduced. Cold hyperalgesia score is significantly increased, and gait is abnormal. The expression levels of serum and trigger points IL-1β, TNF-α, SP, BK, p-IκBα, and p-NF-κB p65, as well as the positive expression intensity of p-NF-κB p65 are significantly increased (P<0.01). Compared with the model group, the positive rate of EMG detection and local convulsion response is significantly reduced in the medium and high dose OK groups (P<0.05). The tenderness threshold and mechanical hyperalgesia threshold increase significantly in the medium and high dose OK groups, and the cold hyperalgesia score is significantly reduced in the high dose OK group (P<0.01). The standing time, swing time, and walking period are significantly increased. The swing speed, maximum contact area, and maximum contact intensity are significantly decreased in the high dose OK group (P<0.05). Moreover, the protein expression levels of p-IκBα/IκBα and p-NF-κB p65/NF-κB p65 are significantly reduced in the medium and high dose OK groups (P<0.05,P<0.01). The positive expression intensity of p-NF-κB p65 is significantly decreased in the high dose OK group (P<0.01).ConclusionThe mechanism of OK in relieving the pain in trigger points of MPS and improving gait abnormalities is related to the downregulation of the NF-κB p65 inflammatory signaling pathway to reduce the expression of inflammatory factors and pain mediators in blood and trigger point tissue.Keywords:myofascial pain syndrome;Osteoking;trigger point;nuclear transcription factor-

Abstract:ObjectiveTo investigate the identification of kidney Yang deficiency syndrome of patients with osteoporosis(OP), and to form the clinical syndrome identification rules of traditional Chinese medicine(TCM).MethodBasic information, etiology, clinical symptoms and other characteristics of 982 OP patients were included, and statistical tests were used to screen the variables associated with kidney Yang deficiency syndrome. Taking the decision tree as the base model, bootstrap aggregation algorithm(Bagging algorithm) was utilized to establish the classification model of kidney Yang deficiency syndrome in OP, generating numerous rules and removing redundancy. Combining least absolute shrinkage and selection operator(LASSO) regression to screen key rules and integrate them to construct an identification model, achieving the identification of kidney Yang deficiency syndrome in OP patients.ResultEighteen key identification rules were screened out, and of these, where 11 rules with regression coefficients>0 correlated positively with the kidney Yang deficiency syndrome, the rule with the highest coefficient was chilliness(present)&feverish sensation over the palm and sole(absent). The other 7 rules with regression coefficients<0 correlated negatively with the syndrome, the rule with the lowest coefficient was reddish tongue(present)&diarrhea(absent)&deficiency of endowment(absent). According to the regression coefficients of each key rule, variables with importance>0.2 were ranked as chilliness, reddish tongue, feverish sensation over the palm and sole, cold limbs, clear urine, diarrhea, deficiency of endowment, prolonged illness. The results of the partial dependence analysis of the identification model showed that compared to OP patients without chilliness, those with chilliness(present) had a 0.266 8 higher probability of being identified as having kidney Yang deficiency syndrome, indicating that this variable had the highest impact on identification of the syndrome. Similarly, compared to OP patients without reddish tongue, those with reddish tongue had a 0.141 9 lower probability of being identified as having kidney Yang deficiency syndrome, indicating that this variable had the highest impact on identifying non-kidney Yang deficiency syndrome. The accuracy, sensitivity, specificity and area under receiver operating characteristic curve(AUC) of the established kidney Yang deficiency syndrome identification model in the test set were 0.865 9, 0.853 7, 0.872 0 and 0.931 5, respectively.ConclusionA precise identification model of OP kidney Yang deficiency syndrome is conducted basing on the rule ensemble method of Bagging combining LASSO regression, and the screened key rules can explain the identification process of kidney Yang deficiency syndrome. In this research, according to the regression coefficients of rules, the importance and partial dependence of variables, combined with the thinking of TCM, the influence of patient characteristics on the identification of syndromes is described, so as to reveal the primary and secondary syndromes of identification and assist the clinical identification of kidney Yang deficiency syndrome.Keywords:osteoporosis;kidney Yang deficiency syndrome;rule ensemble;prediction method;identification model;variable importance;partial dependence251|139|3
<HTML><H-PDF><L-PDF>Updated:2023-11-07- Abstract:ObjectiveTo investigate the efficacy of Huangqintang on mouse models of colitis-associated colon cancer (CAC) and explore the mechanism of Huangqintang in regulating immune function and inflammatory response, inhibiting abnormal cell proliferation, and delaying or inhibiting CAC formation in CAC.MethodC57BL/6J mice were randomly divided into a normal group, model group, mesalazine group, and high- and low-dose Huangqintang groups according to body weight, with 12 mice in each group. Except for the normal group, the rest of the mice were given two intraperitoneal injections of 10 mg·kg-1 azomethane (AOM) and allowed to drink 1.5% dextran sodium sulfate (DSS) freely for seven days and water normally for two weeks. Then, two cycles of ''DSS-drinking water'' were repeated. During the administration of DSS, mice in the normal group and model group were given gavage in equal doses of pure water. Mice in the mesalazine group were given 150 mg·kg-1·d-1 mesalamine suspension for gavage, and mice in the high- and low-dose Huangqintang groups were given 18 and 9 g·kg-1·d-1 Huangqintang for gavage, respectively. Each group was given one dose daily until the end of three cycles. After the intervention, the body weight, colon length, and number of colon tumors in each group were measured, and disease activity index (DAI) scores were performed. The serum contents of interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), interleukin-4 (IL-4), interleukin-10 (IL-10), and gastrointestinal tumor marker carbohydrate antigen-199 (CA199) were detected by enzyme linked immunosorbent assay (ELISA). The colonic lesions were observed by hematoxylin-eosin (HE) staining. The expression of proliferative cell-associated antigen (Ki67) was observed by immunohistochemistry. The expression of T lymphocyte subsets (CD3+, CD4+, CD8+, and CD49b+) in mouse plasma was detected by flow cytometry. Fluorescein isothiocyanate-D (FITC-D) content in mouse serum was detected by fluorescent labeling method. The Western blot method was used to detect the expression of Cyclin D1, cyclin-dependent kinase 2 (CDK2), cyclin-dependent kinase 4 (CDK4), and tightly junction-related Occludin and Claudin-1.ResultCompared with the normal group, the body weight of mice in the model group decreased. DAI score increased significantly, and the colon became shorter. Pro-inflammatory factors such as IL-6, TNF-α, and IL-1β increased, and IL-6 and TNF-α were significantly increased (P<0.05). The inflammatory factor IL-4 (P<0.05) and IL-10 were significantly reduced, and the tumor marker CA199 was significantly increased (P<0.01). HE staining showed that colon lesions, intestinal mucosal epithelial defects with a large number of inflammatory infiltrates, serious crypt structure damage, and glandular arrangement disorder were observed in the model group. Ki67 positive granules were expressed in large areas of colonic tissue. The serum CD4+ and CD4+/CD8+ of mice in the model group decreased significantly (P<0.05), and CD8+ increased significantly (P<0.05). The plasma content of FITC-D in the model group was significantly increased (P<0.05), and the expression of Cyclin D1, CDK2, and CDK4 proteins in colon tissue was significantly increased (P<0.05, P<0.01). In addition, the expression of Occludin and Claudin-1 was significantly decreased. Compared with the model group, the body weight of mice in the mesalazine group and the high- and low-dose Huangqintang groups increased. DAI score decreased, and the colon became longer. IL-6, TNF-α, and IL-1β expression decreased (P<0.05, P<0.01), but there was no significant change in IL-4 and IL-10. The content of CA199 was significantly reduced (P<0.05), and the colomatoid lesions and inflammatory infiltrates were reduced in the mesalazine group and the Huangqintang group. The crypt structure damage was lighter, and the positive expression of Ki67 was reduced. CD4+, CD4+/CD8+, and CD49b+ increased, and the difference was not statistically significant. FITC-D content decreased (P<0.05). The expression of Cyclin D1, CDK2, and CDK4 decreased (P<0.05, P<0.01), and Claudin-1 and Occludin protein expression increased in the high-dose Huangqintang group (P<0.05).ConclusionHuangqintang has a certain delay and inhibitory effect on AOM/DSS-induced inflammatory cancer transformation, and its mechanism of action may be related to regulating immune function and inflammatory response, inhibiting the release of pro-inflammatory factors, repairing damaged intestinal barriers, inhibiting abnormal proliferation of colon cells, and intervening in the formation and development of CAC colon tumors.Keywords:colorectal cancer;inflammation;permeability;proliferation;Huangqintang341|143|7
<HTML><H-PDF><L-PDF>Updated:2023-10-20 - Abstract:Colorectal cancer is one of the common malignant tumors with high morbidity, and changes in lifestyle, dietary structure and environment in China in recent decades have been associated with an increase in the incidence of colorectal cancer. A large number of studies have shown that traditional Chinese medicine(TCM) can be used as a complementary and alternative treatment for colorectal cancer after conventional western medicine treatment. TCM physicians have accumulated a lot of clinical experience in the treatment of patients with stage Ⅰ-Ⅲ colorectal cancer, and have proved that TCM has unique efficacy, but there is still a lack of relevant clinical practice guidelines to standardize and guide the diagnosis and treatment of TCM. Based on this, according to the guideline development process of the World Health Organization Handbook for Guideline Development and the Clinical Evidence Grading Criteria on TCM Based on Evidence Body, under the framework of relevant laws, regulations and technical guidance documents, combined with the evidence of relevant domestic and foreign clinical research in recent years for evidence grading and opinion recommendation, and then the Guidelines for TCM Intervention After Conventional Western Medicine Treatment for Stage Ⅰ-Ⅲ Colorectal Cancer were developed by expert consensus. <HTML><H-PDF><L-PDF>Updated:2023-10-13 - Abstract:ObjectiveTo observe the effect of modified Gegen Qinliantang on the expression levels of proteins related to the farnesoid X receptor/small heterodimer partner/peroxisome proliferator-activated receptor α (FXR/SHP/PPARα) signaling pathway in the liver tissue of db/db model mice with type 2 diabetes mellitus (T2DM) and explore the underlying mechanism of action of modified Gegen Qinliantang.MethodThirty db/db mice were randomly divided into model group, metformin group (0.2 g·kg-1), and high-, medium-, and low-dose modified Gegen Qinliantang groups (31.9, 19.1, 6.4 g·kg-1), with 6 mice in each group. An additional six m/m mice were assigned to the blank group. Respective drugs were administered via oral gavage for 12 weeks. Mouse body weight, fasting blood glucose (FBG), total cholesterol (TC), triglyceride (TG), high-density lipoprotein cholesterol (HDL-C), and low-density lipoprotein cholesterol (LDL-C) levels were measured. Oil red O staining was used to observe hepatic lipid accumulation and periodic acid-schiff (PAS) staining was used to assess hepatic glycogen deposition. Ammonium ferric sulfate staining was used to observe cholesterol deposition in intestinal tissues. Western blot was employed to detect the expression of FXR, cholesterol 7α-hydroxylase (CYP7A1), SHP, and PPARα proteins in liver tissues, and enzyme-linked immunosorbent assay (ELISA) was used to measure serum free fatty acid (FFA) levels.ResultAt the end of the treatment, compared with the blank group, the model group exhibited significant increases in mouse body weight, FBG, FFA, TC, TG, and LDL-C levels (P<0.01), along with significant hepatic lipid droplets, reduced hepatic glycogen, noticeable cholesterol accumulation in intestinal tissues, significantly decreased expression of FXR, SHP, PPARα proteins, and significantly increased expression of CYP7A1 protein in liver tissues (P<0.01). Compared with the model group, the metformin group and the high- and medium-dose modified Gegen Qinliantang groups demonstrated significant reductions in mouse body weight, FBG, FFA, TC, TG, LDL-C levels (P<0.05, P<0.01), significant increases in HDL-C levels (P<0.05, P<0.01), decreased hepatic lipid accumulation, increased hepatic glycogen, reduced intestinal cholesterol accumulation, significantly increased expression of FXR, SHP, PPARα proteins, and significantly decreased expression of CYP7A1 protein in liver tissues (P<0.01).ConclusionModified Gegen Qinliantang may regulate the FXR/SHP/PPARα signaling pathway to suppress FFA levels and improve lipid metabolism in T2DM mice.Keywords:type 2 diabetes mellitus;modified Gegen Qinliantang;farnesoid X receptor/small heterodimer partner/peroxisome proliferators-activated receptor

<HTML><H-PDF><L-PDF>Updated:2023-09-21 - Abstract:In contemporary clinical practice of traditional Chinese medicine (TCM), there is a strong emphasis on treating chronic difficult diseases and preventing diseases before they occur ("pre-disease" treatment). However, the treatment of critical care medicine has not received as much attention and is even facing a trend of diminishing influence in the field of emergency medicine and critical care. Critical care medicine refers to a class of diseases that are characterized by rapid onset, severity, quick progression, and high mortality rates in clinical practice. The growth and development of renowned TCM experts throughout history have invariably been honed through practical experiences in managing critical care medicine. Treatise on Cold Damage (《伤寒论》) is a dedicated work for treating critical care medicine, and the classic prescriptions in Treatise on Cold Damage are particularly central to the treatment of these diseases. In the current clinical context, it is important to break through the prevailing mindset that the strengths of TCM lie solely in treating chronic diseases and maintenance-type illnesses, identify the entry point of TCM in the clinical advantages in the field of critical care medicine, explore the interdisciplinary relationship between Treatise on Cold Damage and critical care medicine, enhance the ability to treat critical care medicine using classic prescriptions, establish a disciplinary system for prescription-based treatment of critical care medicine, reveal clinical evidence for prescription-based treatment of these diseases, and elevate the contribution of TCM in the domain of critical care medicine. These have become a strong call and demand in contemporary TCM clinical practice. Regarding key scientific issues, the system of using classic prescriptions to treat critical care medicine encompasses traditional core theories such as the nature of cold damage, the essence of the six meridians, the implications of specific clauses, and the dosages of classic prescriptions. In the context of future academic inheritance and system construction, it is recommended to focus on clinical challenges, break away from the current limitations of traditional classical disciplinary systems, such as their singular patterns and weak innovative and translational capabilities, suggest defining diseases in critical care medicine responding specifically to TCM, advocate a research model based on combining disease mechanisms with pathology and combining medicinal properties with pharmacology, establish a comprehensive multidisciplinary thinking mode, and encourage collaborative innovative research among medical, educational, research, and manufacturing institutions. This will help chart the path and address key aspects of constructing a comprehensive system for prescription-based treatment of critical care medicine.Keywords:classical prescriptions;

Sixty male C57BL/6J mice were randomized into normal, model, mesalazine (0.3 g·kg-1), and high-, medium-, and low-dose (12.35, 8.22, 4.11 g·kg-1, respectively) Renshen Baidusan groups (n=10). The mice were administrated with corresponding drugs by gavage for 7 consecutive days. The colon tissue was stained with hematoxylin-eosin (HE) to reveal the pathological changes, and Alcian blue-Periodic acid Scheff (PAS/AB) staining was employed to observe the goblet cell changes. The fluorescence expression of reactive oxygen species (ROS) in the colon tissue was detected by the immunofluorescence assay. The activity of superoxide dismutase (SOD) and the content of malondialdehyde (MDA) were measured by the biochemical methods. Western blot was employed to determine the expression levels of proliferating cell nuclear antigen (PCNA), microtubule-associated protein 1 light chain 3 (LC3), leucine-rich repeat-containing G protein-coupled receptor 5 (LGR5), and p62.ResultDestroyed mucosal epithelial structure, intestinal gland destruction, loss of goblet cells, and massive infiltration of inflammatory cells appeared in the model group. Compared with the normal group, the model group showed increased tissue damage injury (TDI) score of the colon tissue, decreased SOD activity and LC3Ⅱ/Ⅰ, PCNA value, and elevated levels of p62, MDA, ROS, and LGR5 (P<0.05). Compared with the model group, different doses of Renshen Baidusan decreased the TDI score, promoted the generation of new goblet cells, elevated the levels of PCNA, LGR5, SOD, and LC3Ⅱ/Ⅰ, and lowered the levels of p62, MDA, and ROS (P<0.05). <HTML><H-PDF><L-PDF>Updated:2023-09-08 - Abstract:ObjectiveTo explore the mechanisms of internal treatment (Renshen Baidusan), external treatment (Yurui Enema), and combination of the two methods in treating intestinal mucosal injury in the rat model of ulcerative colitis (UC) from the changes of phosphatidylinositol-3 kinase (PI3K)/protein kinase B (Akt)/nuclear factor-κB (NF-κB) pathway.MethodFifty SPF-grade SD rats were randomized into blank, model, Renshen Baidusan (15.6 g·kg-1), Yurui Enema (25 g·kg-1), and combined treatment (15.6 g·kg-1 Renshen Baidusan + 25 g·kg-1 Yurui Enema) groups (n=10). The rat model of UC was established in other groups except the blank group by 2,4, 6-trinitrosulfonic acid (TNBS)/ethanol. The rats were administered with corresponding drugs once a day for 14 consecutive days since the 8th day after modeling. The histopathological changes of colon were observed by hematoxylin-eosin (HE) staining. Enzyme-linked immunosorbent assay (ELISA) was employed to measure the levels of tumor necrosis factor (TNF)-α, interferon (IFN)-γ, interleukin (IL)-4, and IL-10 in the colon tissue. The apoptosis of colon epithelial cells was detected by terminal deoxynucleotidyl transferase-mediated nick end labeling (TUNEL). The location and expression of B-cell lymphoma-2 (Bcl-2), Bcl-2-associated X protein (Bax), TNF-α, and IL-6 in the colon tissue were examined by immunohistochemistry. Real-time quantitative fluorescence polymerase chain reaction (Real-time PCR) and Western blot were employed to determine the mRNA and protein levels, respectively, of the proteins in the PI3K/Akt/NF-κB pathway in the colon tissue.ResultIn the model group, HE staining showed a large number of inflammatory cell infiltration in the mucosa and submucosa. Compared with the blank group, the model group showed elevated levels of TNF-α and IFN-γ and lowered levels of IL-4 and IL-10 in the colon tissue, increased apoptosis rate of colon epithelial cells, increased positive expression of Bax, TNF-α, and IL-6, and decreased positive expression of Bcl-2 (P<0.05). Moreover, the model group showed up-regulated mRNA levels of PI3K, Akt, and NF-κB and protein levels of PI3K, p-PI3K, Akt, p-Akt, p65, p-p65, Bax, and cleaved Caspase-3, increased Bax/Bcl-2 and cleaved Caspase-3/Caspase-3 ratios, and down-regulated protein levels of NF-κB suppressor protein α(IκBα), Bcl-2, and Caspase-3 in the colon tissue (P<0.05). Compared with the model group, the internal treatment, the external treatment, and the combination (referred to as the three groups) alleviated the colonic mucosal injury, lowered the levels of TNF-α and IFN-γ and elevated the levels of IL-4 and IL-10 in the colon tissue, decreased the apoptosis rate of colon cells, inhibited the positive expression of Bax, TNF-α, and IL-6, and promoted the positive expression of Bcl-2 (P<0.05). Furthermore, the combination group down-regulated the mRNA level of PI3K (P<0.05). The three groups down-regulated the mRNA levels of Akt and NF-κB and the protein levels of p-PI3K, Akt, p-Akt, p65, p-p65, Bax, and cleaved Caspase-3 in the colon tissue, decreased the Bax/Bcl-2 and cleaved Caspase-3/Caspase-3 ratios, and up-regulated the protein levels of IκBα, Bcl-2, and Caspase-3 (P<0.05).ConclusionRenshen Baidusan, Yurui Enema, and their combination may inhibit the activation of PI3K/Akt/NF-κB signaling pathway and regulate the expression of genes and proteins related to this pathway to achieve anti-inflammatory and anti-apoptotic effects, thus restoring the intestinal mucosal barrier function of UC rats.Keywords:Renshen Baidusan;Yurui Enema;combined internal and external treatment;ulcerative colitis;phosphatidylinositol-3 kinase (PI3K)/protein kinase B (Akt)/nuclear factor-

<HTML><H-PDF><L-PDF>Updated:2023-09-08 - Abstract:ObjectiveTo study the mechanism of Renshen Baidusan in regulating adenylate-activated protein kinase (AMPK)/Unc-51-like kinase 1 (ULK1) autophagy pathway to inhibit mucosal barrier damage in the mouse model of ulcerative colitis (UC).MethodSixty SD rats were randomized into normal, model, sulfasalazine enteric-coated tablets (0.312 5 g·kg-1, western medicine), and high-, medium-, and low-dose (31.2, 15.6, 7.8 g·kg-1, respectively) Renshen Baidusan groups. The UC model was induced by 2,4,6-trinitrobenzenesulfonic acid (TNBS)/50% ethanol. The drugs were administrated by gavage for 2 weeks, and then the histopathological changes of the colon were examined. Real-time quantitative polymerase chain reaction was conducted to measure the mRNA level of AMP-activated protein kinase subunit alpha (AMPKα). Western blot was employed to determine the protein levels of closure protein (Occludin), compact linking protein-2 (Claudin-2), autophagy marker p62, microtubule-associated protein 1 light chain 3B (LC3B), phosphorylated AMPK (p-AMPK), and phosphorylated ULK1 (p-ULK1).ResultCompared with the normal group, the model group showed increased colon injury score (P<0.05), down-regulated mRNA level of AMPKα (P<0.05) and protein levels of p-AMPK, p-ULK1, and Occludin, decreased LC3Ⅱ/Ⅰ ratio (P<0.05), and up-regulated protein levels of p62 and Claudin-2 (P<0.05). Compared with the model group, all the doses of Renshen Baidusan lowered the colon injury score, up-regulated the mRNA level of AMPKα and the protein levels of p-AMPK, p-ULK1, and Occluding, increased LC3Ⅱ/Ⅰ ratio, and down-regulated the protein levels of p62 and Claudin-2. Moreover, the medium-dose group showed a significant intervention effect (P<0.05).ConclusionRenshen Baidusan can protect the intestinal mucosal barrier from damage, and the medium dose showed the best efficacy.
