目的

探讨浓缩当归丸改善卵巢功能不全大鼠卵巢氧化应激损伤的作用及机制。
方法

36只成年雌性SD大鼠,随机分为正常组、模型组、雌二醇(E
2
)/地屈孕酮片组、浓缩当归丸高、中、低剂量组,每组6只。除正常组外,其余各组大鼠每日腹腔注射乙烯基环己烯(VCD)80 mg·kg
-1
造模,连续14 d。从第15天起,模型组灌胃生理盐水2 mL·kg
-1
;雌二醇/地屈孕酮片组灌胃芬吗通0.3 mg·kg
-1
;浓缩当归丸高、中、低剂量组灌胃浓缩当归丸2.08、4.16、8.32 g·kg
-1
,每日1次,连续给药28 d。测定大鼠卵巢指数;酶联免疫吸附测定法(ELISA)检测血清中相关激素E
2
、促卵泡激素(FSH)、卵泡刺激素(FSH)、促黄体生成素(LH)和抗缪勒管激素(AMH)水平,以及血清过氧化物歧化酶(SOD)活性、谷胱甘肽(GSH)含量;硫代巴比妥酸(TAB)法检测血清丙二醛(MDA)含量;苏木素-伊红(HE)染色法观察卵巢形态学变化;免疫组化法(IHC)检测卵巢组织中核因子E
2
相关因子2(Nrf2)、血红素氧合酶-1(HO-1)、SOD2、SOD1的表达;蛋白免疫印迹法(Western blot)检测卵巢组织中Nrf2、HO-1、SOD2、SOD1蛋白的相对表达量。
结果

与正常组比较,模型组大鼠卵巢中生长卵泡明显减少、卵泡颗粒层排列松散;大鼠体质量、卵巢指数和血清AMH、E

水平显著下降,LH、FSH水平均显著升高(

浓缩当归丸能够调节血清激素水平,且增加卵巢组织中Nrf2、HO-1、SOD2、SOD1因子的表达,提高卵巢组织抗氧化能力抵抗氧化应激损伤,从而改善卵巢储备功能。
Objective

To investigate the effect and mechanism of Nongsuo Dangguiwan in improving ovarian oxidative stress in rats with ovarian dysfunction.
Method

Thirty-six adult female SD rats were randomly divided into normal group, model group, positive drug group (Femoston, 0.3 mg·kg
-1
), and high-, medium-, and low-dose groups of concentrated Nongsuo Dangguiwan (2.08, 4.16, 8.32 g·kg
-1
), with six rats in each group. Rats, except for those in the normal group, were injected with 80 mg·kg
-1
vinyl cyclohexene dioxide (VCD) per day for 14 consecutive days to induce ovarian dysfunction. From the 15th day, rats were treated with corresponding drugs by gavage, while those in the model group received 2 mL·kg
-1
saline, once daily for 28 consecutive days. The ovarian index, levels of related hormones including estradiol (E
2
), follicle-stimulating hormone (FSH), luteinizing hormone (LH), and anti-mullerian hormone (AMH) in serum, as well as superoxide dismutase (SOD) activity and glutathione (GSH) content in serum were measured by enzyme-linked immunosorbent assay (ELISA). The malondialdehyde (MDA) content in serum was detected by the thiobarbituric acid (TAB) method. Ovarian morphology was observed by hematoxylin-eosin (HE) staining. The expression of nuclear factor E
2
-related factor 2 (Nrf2), heme oxygenase-1 (HO-1), SOD2, and SOD1 in ovarian tissues was detected by immunohistochemistry (IHC) and Western blot.
Result

Compared with the normal group, the model group showed a significant reduction in growing follicles in the ovary, loose arrangement of granulosa cells in the follicle, decreased body weight, ovarian index, and serum AMH and E
2
levels, increased LH and FSH levels (

0.01), reduced levels of SOD and GSH in serum (

0.01), and increased MDA level (

0.01). Compared with the model group, the groups with drug intervention showed increased ovarian index (

Conclusion

Nongsuo Dangguiwan can regulate serum hormone levels, increase the expression of Nrf2, HO-1, SOD2, and SOD1 in ovarian tissues, and improve ovarian antioxidant capacity to resist oxidative stress injury, thereby improving ovarian reserve function.
