Resümee

Ziel der vorliegenden Studie war es, die Haupt-Chemikalien von AMD zu identifizieren und die Wirkmechanismen von AMD bei der Regulation des zirkadianen Rhythmus von Schlaflosigkeitsratten durch den ERK1/2/MNK/eIF4E-Signalweg zu erforschen. Die Hauptchemikalien von AMD wurden durch hochleistungsfähige Flüssigkeitschromatographie in Kombination mit linearer Ionenfallen-Orbitrap-Massenspektrometrie (UPLC-LTQ/Orbitrap/MS) unter Verwendung von Referenzstandards identifiziert. Sechzig SD-Männchen wurden zufällig einer Leergruppe, einer Modellgruppe, einer Melatonin-Gruppe, einer niedrigen, mittleren und hohen Dosisgruppe von AMD mit jeweils zehn Ratten pro Gruppe zugewiesen, mit Ausnahme der Leergruppe, die intraperitoneal mit 4-Chlorsäure-phenylacetonitril injiziert wurde, um Schlaflosigkeit zu induzieren. Offenes Feldexperiment und zirkadiane Rhythmusstudien wurden durchgeführt, um den Aktivitäts-Ruhe-Rhythmus der Ratten zu messen, HE-Färbung und Nissl-Färbung wurden verwendet, um Veränderungen in der neuronalen Struktur im Hypothalamus zu beobachten, Immunfluoreszenz, Echtzeit-PCR und Westernblot wurden durchgeführt, um die Expression von RNAs und Proteinen im Zusammenhang mit dem ERK1/2/MNK/eIF4E-Signalweg im Hypothalamus zu untersuchen. Im Vergleich zur Leergruppe zeigte die Modellgruppe eine signifikante Zunahme der Gesamtstrecke, der Durchschnittsgeschwindigkeit, der Aufenthaltsdauer im Zentrum und der aufrechten Körperhaltungszeit (P<0,01), sowie eine signifikante Verlängerung der Gesamtaktivitäts- und Ruhezeit (P<0,01); eine verstreute hypothalamische neuronale Anordnung, eine Abnahme der Neuronenzahl, das Verschwinden einiger Zellkerne und die Zerstörung von Nukleolen, sowie einen signifikanten Anstieg des Zellapoptoseindexes (P<0,01); ein trübes Zytoplasma, eine hellere Farbe des Nissl-Körpers und eine signifikante Zunahme des Nissl-Körperapoptoseindexes (P<0,01); eine signifikante Abnahme der ERK1/2-, MNK- und eIF4E-RNA-Expression im Hypothalamus (P<0,01); eine signifikante Abnahme der Proteine ERK1/2, phosphoryliertes (p)-ERK1/2, MNK, phosphoryliertes (p)-MNK, eIF4E und phosphoryliertes(p)-eIF4E im Hypothalamus (P<0,01). Im Vergleich zur Modellgruppe zeigte die Hochdosisgruppe von AMD eine signifikante Abnahme der Gesamtstrecke, der Durchschnittsgeschwindigkeit, der Aufenthaltsdauer im Zentrum und der aufrechten Körperhaltungszeit (P<0,01), ähnlich der Melatoningruppe. Die Gesamtstrecke, die Durchschnittsgeschwindigkeit und die aufrechte Körperhaltungszeit in den mittleren und niedrigen Dosisgruppen von AMD zeigten eine signifikante Abnahme (P<0,01). Die Gesamtaktivitätszeit und die Gesamtaktivitätszeit wurden in allen AMD-Dosierungsgruppen signifikant reduziert (P<0,01), und die Gesamtaktivitätszeit in der Hochdosisgruppe von AMD wurde signifikant verlängert (P<0,01). Fazit: AMD enthält fünfzig Chemikalien, wie Flavonoide, Phenylpropanoide, Triterpenoidsaponine, die den Vorteil haben, Schlaflosigkeit durch die synergetische Wirkung mehrerer Komponenten und Wege zu verbessern, indem sie die positive Regulation der Proteine im Zusammenhang mit dem ERK1/2/MNK/eIF4E-Signalweg zur Verbesserung des gestörten zirkadianen Rhythmus von Schlaflosigkeitsratten induziert durch PCPA erhöhen.
