Ziel ist es, die chemische Zusammensetzung des Dan-Shen-Getränks und die Blutkomponenten nach oraler Verabreichung bei Ratten zu erläutern. Methode: Verwendung von Ultra-Hochleistungs-Flüssigkeitschromatographie gekoppelt mit Quadrupol-Flugzeit-Tandemmassenspektrometrie (UPLC-Q-TOF-MS/MS) in Kombination mit der PeakView 1.2-Software. Basierend auf Retentionszeit, Quasi-Molekularionenpeak, sekundären Fragmentionen und Literaturabgleich wurde eine systematische Charakterisierung und Identifikation der Dan-Shen-Wasserextrakte und der nach oraler Gabe im Blut auftretenden Komponenten bei Ratten durchgeführt. Weiterhin wurden die chemischen Bestandteile von gemeinsam mit Sandelholz und Amomum gekochten Dan-Shen-Wasserextrakten bestimmt, um die Herkunft der chemischen Bestandteile im Dan-Shen-Auszug klarzustellen. Ergebnisse: Insgesamt wurden 73 Verbindungen im Dan-Shen-Wasserextrakt identifiziert, darunter 21 Phenolsäuren, 23 Diterpene, 6 Flavonoide, 7 organische Säuren, 3 flüchtige Öle und 13 andere Typen. In der Arzneimittel-haltigen Rattenplasma wurden 36 Proto-Komponenten, die in das Blut gelangten, und 15 Metaboliten identifiziert. Die Hauptstoffwechselwege umfassen Reduktion, Hydratation, Hydroxylierung, Demethylierung, Methylierung und Sulfatierung. Diese Metaboliten stammen hauptsächlich von Danshenon- und Phenolsäurekomponenten. Fazit: Die Blutkomponenten des Dan-Shen-Wasserextrakts bestehen hauptsächlich aus Phenolsäuren und Danshenon-Komponenten, was möglicherweise die pharmakologische Wirkstoffbasis darstellt und als Referenz für Wirksamkeitsforschung, Qualitätskontrolle und klinische Anwendung des Dan-Shen-Getränks dienen kann.
