Ziel: Danggui ist ein häufig verwendetes Arznei- und Nahrungsmittel mit häufigen Marktverfälschungen, die die klinische Wirksamkeit von Zubereitungen stark beeinträchtigen. Derzeit sind qualitative Nachweistechniken für Verfälschungen relativ ausgereift, quantitative Nachweismethoden für Verfälschungen in verarbeiteten Produkten fehlen jedoch. In dieser Studie wurde am Beispiel von Danggui und seiner leicht verwechselbaren nahen Verwandten „Bodendanggui“ (koreanischer Danggui) eine quantitative Nachweismethode entwickelt, um eine molekulare quantitative Nachweismethode (Herb-Q) für Danggui und seine verarbeiteten Produkte zu etablieren und damit eine Demonstration für die Entwicklung quantitativer Nachweistechniken für Danggui-Rohstoffe und zugehörige Gesundheitsprodukte zu bieten. Die Methode basiert auf der Auswahl spezifischer Einzelnukleotid-Polymorphismen (SNP) im Chloroplastengenom von Danggui und koreanischem Danggui anhand der vollständigen Chloroplastengenomsequenz. Für die quantitativen Untersuchungen wurden danggui-spezifische Stellen ausgewählt und die Methodik unter Verwendung von Kräuterpulvermischungen mit unterschiedlichen Verfälschungsanteilen geprüft (Linearität, Quantifizierungsgrenze, Nachweisgrenze und Reproduzierbarkeit). Die Genauigkeit der Herb-Q-Methode wurde mit drei verschiedenen Verfälschungsgraden (15 %, 25 %, 35 %) in einem durchblutungsfördernden Schmerzmittelpulver validiert. Ergebnisse: Durch den Vergleich von 123 Chloroplastengenomsequenzen der Gattung Danggui wurden unter Berücksichtigung der innerartlichen Konservierung, artspezifischen Eigenschaften und der Anforderungen an das Pyrosequencing zwei Stellen im Chloroplastengenom ermittelt: Position 9674 (A/G) in NC_042826.1 und Position 38592 (T/C) in NC_029393.1, die als spezifische molekulare Identifikationsstellen für Danggui bzw. koreanischen Danggui dienen. Die danggui-spezifische Position 9674 (A/G) wurde für die Herbe-Q-Methode ausgewählt. Die erzielte lineare Beziehung hatte einen R²-Wert von 0,9974 (>0,99), was auf eine gute Linearität hinweist. Die Quantifizierungs- und Nachweisgrenzen lagen bei 2,0 %, die Reproduzierbarkeit war mit einem relativen Standardabweichung (RSD) von <2,0 % gut. Das auf der Herb-Q-Methode basierende Quantifizierungssystem ermittelte eine mittlere Abweichung von 1,3 % bei der dreifachen molekularen Quantifizierung des Verfälschungsanteils koreanischen Danggui im durchblutungsfördernden Schmerzmittelpulver. Fazit: Diese Studie zeigt, dass die auf Position 9674 (A/G) des Chloroplastengenoms NC_042826.1 basierende Herb-Q-Quantifizierungsmethode gut geeignet, objektiv und genau für Danggui und „Bodendanggui“ sowie deren verarbeitete Produkte ist und eine technische Unterstützung für die quantitative Nachweistechnik kommerzieller Danggui-Derivate wie chinesische Arzneimittel, Nahrungsergänzungsmittel und Gesundheitsprodukte bietet.
