Ziel ist die Erforschung des Wirkmechanismus der Jianpi Xiaoaifang-Formel, die die induzierbare Stickstoffmonoxid-Synthase-Arginase 1 (iNOS-ARG1) Stoffwechselachse reguliert, um eine mittels mitochondrialer reaktiver Sauerstoffspezies (mito-ROS) vermittelte mitochondriale Struktur- und Funktionsstörung zu induzieren und dadurch das Fortschreiten von Kolonkarzinomen zu hemmen. Ein Modell zur Störung des Arginin-Metabolismus in menschlichen Kolonkarzinomzellen HCT116 wurde durch die gemeinsame Behandlung mit rekombinantem humanem Interferon-γ (IFN-γ, 10 μg·L⁻¹) und N-(ω)-Hydroxy-L-Arginin (Nor-NOHA, 200 μmol·L⁻¹) für 24 Stunden aufgebaut, gefolgt von Interventionen mit 5 %, 10 % und 20 % Jianpi Xiaoaifang enthaltendem Arzneimittelserum. Im Vergleich zur Kontrollgruppe zeigte die Modellgruppe eine signifikante Hochregulierung von iNOS, Herunterregulierung von ARG1, Abnahme von ARG1-iNOS, verringerte Gehalte an L-Arginin und Harnstoff sowie erhöhte Citrullinwerte (P<0,05); gleichzeitig war die Anhäufung von mito-ROS signifikant erhöht, das JC-1 Rot/Grün-Verhältnis vermindert, es traten mitochondriale Schwellungen und Kristabrüche auf, was darauf hinweist, dass ein metabolischer Ungleichgewicht mitochondriale Schäden verursachen kann. Im Vergleich zur Modellgruppe senkten alle Dosierungen von Jianpi Xiaoaifang die ARG1/iNOS weiter ab, erhöhten die Anhäufung von Stickstoffmonoxid (NO) und reaktiven Stickstoffspezies, verringerten zusätzlich die L-Arginin- und Harnstofflevel und erhöhten signifikant das Citrullin (P<0,01); die EdU-Positivrate, Klonbildungsrate, Wundheilungsrate und Anzahl der Transwell-durchdringenden Zellen nahmen mit steigender Konzentration der Arzneimittelserumsignifikant ab (P<0,01); der mito-ROS-Level stieg, das JC-1 Rot/Grün-Verhältnis sank weiter. TEM zeigte eine verstärkte mitochondriale Schwellung und Vakuolisierung, eine Herunterregulierung des MFN2-Proteins und eine Hochregulierung von DRP1 (P<0,01) in dosisabhängiger Weise. Fazit: Die Jianpi Xiaoaifang-Formel kann auf der Grundlage eines gestörten Arginin-Stoffwechsels den NO-vermittelten oxidativen und nitrosativen Stress weiter aktivieren, die Akkumulation mitochondrialer ROS, Membranpotentialkollaps und Dynamikunbalance induzieren, die Proliferation und Migration von Kolonkarzinomzellen hemmen und so einen synergistischen "Metabolismus-Mitochondrien"-gerichteten Anti-Krebs-Mechanismus offenlegen.
