卵巢癌是一种发病率和病死率高的女性生殖系统恶性肿瘤,目前卵巢癌的治疗主要是以初次肿瘤细胞减灭术联合紫杉醇/卡铂(TC)化疗方案为主,但5年生存率在36%~46%。中医药作为祖国传统医学,以其独有的药物及用药特点,可以针对卵巢癌发生发展的相关环节和涉及的不同分子靶点发挥着一定的积极作用,其中淫羊藿、黄芩、姜黄、银杏叶、柴胡、金银花等中的黄酮类中药单体成分具有一定的代表性,有大量实验研究证实其具备显著的抗肿瘤活性,已被广泛应用在不同恶性肿瘤的治疗中。回顾文献,通过对多个数据库检索黄酮类中药单体的研究,发现黄酮类中药单体可通过调控卵巢癌的多条信号通路,如磷脂酰肌醇3-激酶(PI3K)/蛋白激酶B(Akt)/哺乳动物雷帕霉素靶蛋白(mTOR)信号通路、Janus激酶(JAK)/信号转导与转录激活因子(STAT)信号通路、丝裂原活化蛋白激酶(MAPK)信号通路、分泌型糖蛋白(Wnt)/
β
-连环蛋白(
β
-catenin)信号通路、Notch信号通路、核转录因子-
κ
B(NF-
κ
B)信号通路等,从而发挥抑制细胞增殖,诱导细胞凋亡,阻滞细胞周期,降低细胞侵袭、迁移能力,抑制肿瘤血管生成,诱导自噬,逆转铂耐药性等作用,以抑制卵巢癌的发生和发展。基于黄酮类中药单体调控信号通路以发挥抗卵巢癌作用方面作一综述,以期为研究黄酮类中药单体抑制卵巢癌的发生发展提供理论依据。
Ovarian cancer is a malignant tumor of female reproductive system with high morbidity and mortality. Currently, ovarian cancer patients are mainly treated by primary debulking surgery combined with taxotere/cyclophosphamide (TC) chemotherapy, with the five-year survival rate of 36%-46%. Chinese medicinal materials play a positive role in preventing the occurrence and development of ovarian cancer via multiple targets. The flavonoid monomers in representative Chinese herbal medicines, such as Epimedii Folium, Scutellariae Radix, Curcumae Longae Rhizoma, Ginkgo Folium, Bupleuri Radix, and Longicerae Japonicae Flos, have been proved to have significant anti-tumor activity and been widely used in the treatment of malignant tumors. We reviewed the relevant literature and summarized that flavonoid monomers can regulate multiple signaling pathways to inhibit cell proliferation, block tumor cell cycle, induce apoptosis and autophagy, reduce the ability of cell invasion and migration, inhibit tumor angiogenesis, and reverse platinum resistance, thereby inhibiting the occurrence and development of ovarian cancer. Such pathways include phosphatidylinositol 3- kinase (PI3K)/protein kinase B (Akt)/mammalian target of rapamycin (mTOR) signaling pathway, Janus kinase (JAK)/signal transducer and activator of transcription 3 (STAT3) signaling pathway, mitogen-activated protein kinase (MAPK) signaling pathway, secreted glycoprotein (Wnt)/
β
-catenin signaling pathway, Notch signaling pathway, and nuclear factor kappaB (NF-
κ
B) signaling pathway. By reviewing the regulatory effect of flavonoid monomers on the signaling pathways of ovarian cancer, we aim to provide a theoretical basis for the research on the roles of flavonoid monomers in inhibiting the occurrence and development of ovarian cancer.
