Objective:To study the anti-invation effct of triptolide for U87 glioma cells and its molecular mechanism.Method: MTT assay was used to determine the growth inhibition by triptolide in glioma U87 cells.Tumor invasion and migration were examined by Transwell chamber assay and wound-healing method.Real-time PCR was conducted to detect alteration of matrix metallopeptidase 9(MMP-9) and Cathepsins B(CB).The alteration of nuclear factor kappa B(NF-κB) p65 expression were studied by Western blot.Result: Triptolide could significantly inhibite glioma U87 cell proliferation
reduce the cell ability of migration and invasion
down-regulating its MMP-9
CB and NF-κB p65 expression level.Conclusion: Triptolide could depress the invasion of glioma cells owing to down-regulating the level of NF-κB pathway which could inhibit MMP-9 activities
reduce the ability of hydrolyzing extracellular matrix in vitro.
