Objective: To optimize purification technology of total flavonoids in Angelica pubescens by macroporous resin. Method: With adsorption rate and desorption rate of total flavonoids as indexes
macroporous resin models were optimized by static adsorption and desorption experiments as well as dynamic adsorption and desorption tests;Purification technology parameters of total flavonoids were optimized by single factor test. Result: LSA-33 macroporous resin presented the best effect of purification
optimum purification conditions were as followings:the concentration of total flavonoids from sample solution 0.1 g·mL-1
pH of sample solution 2-3
adsorption rate 4 BV·h-1
eluted with 3 BV 75% ethanol
desorption rate 3 BV·h-1
collected eluent
purity of total flavonoids in dry extract of A.pubescens was changed from 9.87% to 32.59%. Conclusion: LSA-33 macroporous resin was suitable for preliminary purification of total flavonoids in A.pubescens.
