目的: 观察益智仁挥发油对大鼠快动眼(REM)睡眠剥夺恢复后脑组织氨基酸类神经递质L-谷氨酸(Glu)及γ-氨基丁酸(GABA)含量及其受体表达的影响。 方法: 将Wistar大鼠随机分为空白对照组(等容生理盐水)
REM睡眠剥夺恢复组(等容生理盐水)
阳性对照组(安定0.18 g·kg-1·d-1)
益智仁挥发油低、高剂量组(9
27 g·kg-1·d-1)。各组灌胃给药2周后
除空白对照组外其余各组用自制改良多平台法剥夺大鼠睡眠48 h制作REM睡眠剥夺模型
睡眠恢复6 h后采用高效液相色谱法检测各组大鼠大脑皮质及下丘脑氨基酸类神经递质Glu及GABA含量变化
采用免疫组化染色和mRNA半定量RT-PCR法检测大脑皮质及海马部位γ-氨基丁酸A受体(GABAAR)α1和γ2亚单位的表达变化
并观察益智仁挥发油对这些指标的影响。 结果: 与空白对照组比较
REM睡眠剥夺恢复组大鼠大脑皮质及下丘脑Glu含量显著升高
而GABA含量显著降低(P<0.01)
大脑皮质及海马部位的GABAARα1和γ2亚单位免疫化学累积吸光度明显较低(P<0.01)
皮质部位GABAARα1和γ2 mRNA的表达均显著下调(P<0.01)
经益智仁挥发油干预后各指标明显趋于正常状态改变(P<0.01
P<0.05)。 结论: REM睡眠剥夺恢复可改变皮质及下丘脑部位氨基酸类神经递质Glu

并使皮质及海马部位GABAARα1和γ2亚单位表达明显下调
益智仁的益智健脑作用机制与其调节神经递质含量及上调大脑皮质及海马部位GABAARα1和γ2亚单位的表达有关。
Objective: To observe the effects of volatile oil extracted from Alpinia oxyphylla fructus(VOA) on amino acid neurotransmitters glutamic acid (Glu)
gamma-aminobutyric acid (GABA) and its receptor expression in the brain of the model rats with the rapid eye movement(REM) sleep deprivation recovery. Method: Wistar rats were randomly divided into 5 groups: blank control
sleep deprivation recovery
positive control group
sleep deprivation recovery+high and low-dose VOA treatment. After two weeks of administration
model were established with REM sleep deprivation(SD) induced by maded-self multiple platform method (MMPM). Six hours later
the levels of amino acid neurotransmitters Glu and GABA were detected in cortex and hypothalamus of the model rats by HPLC.Using immunohistochemical staining and semi-quantitative analysis of a retrovirus-polymerase chain reaction (RT-PCR)
γ-aminobutyric acid A receptor (GABAAR) α1 and γ2 subunit expression were detected in cortical and hippocampal parts of the model rats.Whats more
after intervention of VOA
those indicators were studied too. Result: Compared to blank control group
model group rats content of Glu showed a considerable upper
while GABA showed a considerable down
GABAARα1 and γ2 subunit immunochemistry absorbance accumulated were lower significantly (P<0.01) in cortex and hippocampus
and GABAARα1
γ2 mRNA expression showed lower significantly (P<0.01) in cortex.While those indicators tended to the normal range after the treatment of VOA(P<0.01
P<0.05). Conclusion: REM sleep deprivation recovery could regulate amino acid neurotransmitters and make expression of GABAAR α1 and γ2 subunit markedly down in cerebral cortex and hippocampus. VOA can regulate amino acid neurotransmitters to normal state trend
and upward adjusting GABAAR α1 and γ2 subunit expression in the rat brain. This may be one of the mechanisms for VOAs affect of nourishing the brain and improving intelligence.
