Objective: To optimize separation and purification technology of total flavonoids and puerarin from Pueraria lobata by macroporous adsorption resin. Method: Static adsorption and desorption performance of four kinds of different polar macroporous adsorption resins(S-8

HPD-100) for total flavonoids and puerarin from P. lobata were investigated
in order to select optimal resin;With yield and purity of total flavonoids and puerarin as indexes
single factor test was used to investigate adsorption and elution conditions. Result: AB-8 macroporous resin was chosen
its optimized technology conditions were as followings:the concentration of sample solution 1.0 g·mL-1
mass ratio of raw material-resin 1:2
diameter-height ratio of column 1:6
washed by 2 BV water
eluted with 2 BV 80% ethanol
elution flow rate 2 mL·min-1.Elution rate of total flavonoids and puerarin were more than 92%
purity of total flavonoids and puerarin could reach 80% and 25%
respectively. Conclusion: AB-8 macroporous adsorption resin was sutiable for enrichment total flavonoids and puerarin from P. lobata
it was a kind of ideal separation and purification medium
