分别ip利血平(2 mg ·kg-1)或5-羟色氨酸(5-HTP
150 mg ·kg-1)
观察葛根异黄酮对利血平所致小鼠眼睑下垂、体温下降和运动不能的拮抗作用
以及对5-HTP诱导小鼠甩头行为的影响。 结果: 开场实验中
正常组小鼠2 min内穿格次数和前肢上抬次数分别为(57.23±18.35)
(22.16±13.28)次
各给药组与之相比均无显著差异;正常组小鼠FST和TST不动时间分别为(132.7±37)
(98.4±28)s
葛根异黄酮低、中、高剂量均显著缩短了不动时间(P<0.05)
其中100 mg ·kg-1葛根异黄酮组小鼠FST和TST不动时间分别缩至(72.1±27)
(68.8±19)s;葛根异黄酮低、中、高剂量显著增强了5-HTP诱导的小鼠甩头行为
使甩头次数由(4.7±2.3)次分别增至(16.2±7.9)
(27.4±13.1)
(28.8±12.5)次
差异具有显著意义(P<0.001)
且呈剂量依赖性;葛根异黄酮对利血平所致的小鼠眼睑下垂、运动不能以及体温下降等症状亦有显著对抗作用(P<0.05)。 结论: 葛根异黄酮具有显著的抗抑郁作用
其机制可能与调节单胺能神经系统功能有关。
Objective: To investigate the anti-d-epressant effects of pueraria isoflavone in mice. Method: Male KM mice were randomly divided into 5 groups:control groups (normal saline)
fluoxetine group (10 mg ·kg-1)
pueraria isoflavone groups (50

forced swimming test (FST)
tail suspension test (TST)
5-Hydroxytryptophan (5-HTP)-induced head-twitch test and reserpine-antagonism test were carried out to evaluate the anti-depressant effects of pueraria isoflavone. In particular
reserpine (2 mg·kg-1) and 5-HTP (150 mg·kg-1) were given by intrapertioneal injections before the reserpine-antagonism test and 5-HTP-induced head-twitch test respectively. Result: The number of wearing and grooming in 2 min in control mice were (57.23±18.35) and (22.16±13.28) respectively
and there was no significant different between pueraria isoflavone groups and control group. The immobility time of FST and TST in control mice was (132.7±37)
(98.4±28 ) s respectively and there were significant decrease in pueraria isoflavone groups. pueraria isoflavone at dose of 100 mg·kg-1could short immobility time to (72.1±27)
(68.8±19) s. Moreover
pueraria isoflavone enhanced the head-twitch respone evoked by 5-HTP in dose-dependent manner (P<0.001). Compared with model group
pueraria isoflavone could significantly reverse the ptosis
acinesia
and decrease in body temperature induced by reserpine in mice (P<0.05). Conclusion: Pueraria isoflavone has anti-depressant effects
and its mechanism may be related to regulating the functions of monoaminergic nervous system.
