Objective:To optimize extraction technology of quercitrin from bark of Carya cathayensis
and establish its content determination method. Method: Ethanol reflux extraction method was conducted
extraction solution was absorbed by polyamide to remove dopant
then condensed
crystallized and recrystallized to obtain pure product. After identification by NMR
the content of quercitrin was determined by HPLC.Orthogonal test was used to investigate influence of the concentration of ethanol
extraction time and extraction times on yield of quercitrin. Result: NMR verified product as quercitrin
HPLC showed that quercitrin had a good linear relationship in the range of 4.8-91.2 mg·L-1
the content of quercitrin from bark of C. cathayensis was up to 8.37%.Optimum extraction technology was:reflux extracted 2 times with 10 times the amount of 60% ethanol
1 h per time
extraction rate of quercitrin 90%. Conclusion: Established content determination method of quercitrin from C. cathayensis was accurate and simple
optimized extraction technology was stable and feasible.
