摘要

目的:探讨滋补脾阴方药(ZibuPiyinRecipe

ZBPYR)对大鼠脊髓损伤(spinalcordinjury

SCI)的治疗效果以及对低氧诱导因子(hypoxiainduciblefactor

HIF)-1α的表达的影响。方法:将清洁级Wistar大鼠36只随机分为假手术组、阳性对照组(喜得镇组

0.6mg·kg-1)、SCI组和ZBPYR低、中、高剂量组(分别为7

21g·kg-1)。各组大鼠分别于伤后72h处死

并用Western-blot和RT-PCR方法检测HIF-1α和HIF-1αmRNA的表达。结果:伤后72h

HIF-1α和HIF-1αmRNA在SCI组的表达量显著高于假手术组、ZBPYR低、中、高剂量组和阳性对照组

差异具有统计学意义(P＜0.05);HIF-1α和HIF-1αmRNA的表达在ZBPYR低、中、高剂量组间的差异具有统计学意义(P＜0.05)。结论:ZBPYR可改善受损脊髓局部微环境的缺血缺氧程度来抑制SCI后继发性损伤的发生和发展

其作用效果具有浓度依赖性。

Abstract

Objective:To study the effect of Zibu Piyin Recipe (ZBPYR)on spinal cord injury and the influence on expression of hypoxia inducible factor(HIF) -1α. Method:Thirty-six adult Wistar rats were divided into six groups randomly:sham operated group

positive control group(hydergin

0.6 mg·kg-1)

ZBPYR low

middle and high dose group(7

21 g·kg-1)

SCI group. The rats were killed at 72 h post-injury. The expression of HIF-1α and HIF-1α mRNA was detected with RT-PCR and Western-blot assay. Result:At 72 h post-injury

the expression of HIF-1α and HIF-1α mRNA in SCI group were higher than that in the other groups (P＜0.05)

and the expression of HIF-1α and HIF-1α mRNA in ZBPYR high

middle and low dose group were different statistically (P＜0.05). Conclusion:The ZBPYR could improve the function of neurons though improving oxygen-deficiency in local spinal cord tissue after injury.
