摘要

目的: 探讨不同采收月份明党参根中L-天门冬酰胺的动态累积规律。方法: 采用邻苯二甲醛(OPA)柱前衍生HPLC测定明党参中L-天门冬酰胺的含量。取供试品溶液1 mL

OPA衍生试剂1 mL

硼酸缓冲液(0.2 mol·L-1

pH 10.5)5 mL

定容至10 mL

反应时间20 min;流动相:A为1.35%醋酸钠缓冲液(pH 7.3)

B为1.35%醋酸钠(pH 7.2)-乙腈-甲醇(100∶175∶225)。洗脱梯度(0~20 min

100％ B

维持8 min)

柱温 40 ℃

检测波长 333 nm。结果: 在质量浓度100~1 000 mg·L-1对不同采收期的样品在实验条件下测定

L-天门冬酰胺平均含量为0.26%~1.87%。结论: L-天门冬酰胺含量在5~8月及1月较高

9月至次年4月较低。

Abstract

Objective: To study the dynamic accumulative law of L-asparagine in Changium smyrnioides from different collection month. Method: L-asparagine in roots of C. smyrnioides from different collection month were derived with o-phthaldialdehyde (OPA) in pre-column

and separated by RP-HPLC. L-asparagine was determined. 1 mL sample solution

1 mL colorimetric reagent and 5 mL borax buffe (0.2 mol·L-1

pH 10.5) were settled to the volume of 10 mL and reacted 20 min; mobile phase A was 1.35% sodium acetate buffer (pH 7.3)

mobile phase B was 1.35% sodium acetate (pH 7.2)-acetonitri-methanol (100∶175∶225)

0-20 min A∶B (93∶7)

20-25 min A∶B (0∶100)

at the rate of 1 mL·min-1. The temperature of column was 40 ℃ and the wavelength was at 333 nm. Result: The content of L-asparagine was 0.26%-1.87% in 100-1 000 mg·L-1. Conclusion: The contents of L-asparagine was higher in May-August

January than other months.
