Objective: To study the effect of Qize decoction at endoplasmic reticulum stress(ERS) in rats with chronic renal failure(CRF). Method: SD rats were fed with adenine of 2.5% to establish chronic renal failure model. They were divided into five groups:normal group
model group
Niaoduqing group(2.5 g·kg-1·d-1)
Qize decoction of high dosage(50.4 g·kg-1·d-1)group and Qize decoction of low dosage(12.6 g·kg-1·d-1) group
each group has 10 rats. All groups were infused with adenine into stomach in addition to normal group
at the same time
each rat in chinese herbal compound group was also infused with chinese herbal compound into stomach and Niaoduqing group was given Niaoduqing liquid for 24 days. Body weight was observed dynamically
blood urea nitrogen(BUN) and serum creatinine(SCr) levels were checked by automatic biochemical analyzer.The apoptosis of renal was evaluated by means of terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling(TUNEL).The expression of C/EBP-homologous protein(CHOP) and Caspase-12 was detected by Western bloting. Result: Compared with the control group BUN(4.19±0.77)mmol·L-1 and SCr(15.89±2.62)μmol·L-1
in the model group BUN(17.21 ±4.63) mmol·L-1 and SCr levels (67.00 ±25.62) μmol·L-1 were much higher(P<0.01).The results of TUNEL showed that the few numbers of apoptosis in renal tubular epithelial cells in normal group were found
but more in model group. The expression of CHOP and Caspase-12(P<0.01)were much higher in the model group. Compared with the model group
the number of apoptosis in renal tubular epithelial cells and the expression of CHOP and Caspase-12 were reduced(P<0.05) in Qize decoction low dosage group and Niaoduqing group. Conclusion: Qize decoction low dosage could protect the adenine-induced renal damage
maybe related to that it can inhibit the high expression of ERS-related apoptosis-molecule CHOP and Caspase-12.
