目的: 研究绿原酸对四氯化碳(CCl4)所致肝损伤小鼠的保护作用。 方法: 将60只昆明种小鼠随机分为6组
各组每天ig给药容量为20 mL·kg-1·d-1
连续10 d。正常组和模型组给予蒸馏水
阳性组给予联苯双酯0.60 g·kg-1
绿原酸高、中、低剂量组分别给予绿原酸14

3.5 mg·kg-1。末次给药后禁食不禁水
8 h后
除正常组外
其余各组均ip 0.08% CCl4花生油溶液10 mL·kg-1
12 h后眼球取血
分离血清
检测血清中的谷丙转氨酶(ALT)和谷草转氨酶(AST)
同时测定肝匀浆中的超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)的活性和丙二醛(MDA)的水平。将肝大叶HE染色
观察绿原酸对小鼠肝脏组织病理学的影响。 结果: 绿原酸14

3.5 mg·kg-1剂量组小鼠的肝指数依次为(2.00±1.16)
(2.40±1.52)
(3.20±1.09)%
脾指数为(0.29±2.61)
(0.36±0.19)
(0.49±1.21)%;小鼠血清中ALT
AST活性依次分别为(40.01±7.14)
(51.19±9.45)
(62.66±9.01)U·L-1
(43.59±4.32)
(52.99±5.97)
(66.97±7.34)U·L-1
小鼠肝组织中MDA 的水平依次为(21.68±10.28)
(28.54±6.84)
(35.51±9.87) nmol·g-1
均明显低于模型组
差异有统计学意义(P<0.01)。绿原酸14

小鼠肝组织中SOD活性依次为(112.98±8.41)

均明显高于模型组
差异有统计学意义(P<0.01)。同时
绿原酸各剂量组肝组织病变程度明显减轻
肝细胞再生明显
偶伴有点状坏死
汇管区少量炎细胞浸润。 结论: 绿原酸具有显著的抗肝损伤的作用
其作用可能与其能清除体内自由基和抗氧化的作用有关。
