Objective: To study on the influence and mechanisms of N-Maleoyl-L-Valine Curcumin Ester (MVC) on MGC-803 cell cycle. Method :The role of MVC on cell growth was tested in vitro by MTT. Cell cycle was analyzed by flow cytometry. The level of the protein involved in cell cycle arrest metabolism was detected by western blotting. Result :MCV markedly inhibidited MGC-803 cell growth and significantly blocked cell cycle progression in the S and G2/M phase in MGC-803 cells. The expression of p21WAF1/CIP1 was increased
and while expression of Cdc 2 was decreased in MGC-803 cell treated with 20 μmol ·L-1 MCV for 24 hour. Conclusion : MCV induces S and G2/M arrest in MGC-803 cells via a mechanism linked with up- regulation of p21WAF1/CIP1and dowh-regulation of Cdc 2.
