目的: 观察丹蒌片对异丙肾上腺素(isoproterenol
ISO)致急性心肌梗死(acute myocardial infarction
AMI)大鼠心肌梗死面积和基质金属蛋白酶-2(matrix metalloproteinase-2
MMP-2)、基质金属蛋白酶-9(matrix metalloproteinase-9
MMP-9)及基质金属蛋白酶抑制剂-1(tissue inhibitors of metalloproteinases-1
TIMP-1)蛋白表达的影响。 方法: 雄性Wistar大鼠80只随机分为4组:空白组(生理盐水10 mL ·kg-1
n=20)、模型组(生理盐水10 mL ·kg-1
n=20)、丹蒌高剂量组(900 mg ·kg-1
n=20)、丹蒌低剂量组(450 mg ·kg-1
n=20)
每组大鼠再随机配对分成10小组
连续ig 14 d。14 d后除空白组以外
其他各组一次性sc ISO 15 mg ·kg-1制备急性心肌梗死模型。造模后1 d
7 d观察心肌病理形态学、梗死面积、心肌MMP-2、MMP-9及TIMP-1蛋白表达水平。 结果: 与模型组比较
丹蒌片900 mg ·kg-1可明显减轻缺血心肌病理损害程度
缩小心肌梗死面积
降低缺血心肌MMP-2(A依次为42.10±6.58及26.71±6.84
P<0.01)
MMP-9(A依次为43.37±8.78及32.91±5.21
P<0.01)蛋白表达
对TIMP-1表达无明显影响。 结论: 丹蒌片可能通过抑制MMP-2
MMP-9蛋白表达
减少心肌梗死面积
促进梗死愈合
防治早期心室重构(ventricular remodeling
VR);丹蒌片对TIMP-1表达无明显影响
对MMP活性的抑制作用可能不通过TIMP实现。
Objective: To study the effect of Danlou tablet on myocardial Infarct size and MMP-2
MMP-9 and TIMP-1 protein expression in rats with acute isoproterenol-induced myocardial infarction. Method :Eighty male Wistar rats were randomly divided into 4 groups:① blank control group(normal saline 10 mL ·kg-1
n=20);② model control group(normal saline 10 mL ·kg-1
n=20)③Danlou large dose group(900 mg ·kg-1
n=20);④Danlou small dose group(450 mg ·kg-1
n=20).Each group of rats were randomly paired into 10 pairs
continuously ig 14 d. Except the blank control group
acute myocardial infarction models were induced by subcutaneous injection of Iso 15 mg ·kg-1 after 14 days of drug treatment. Pathological changes in myocardial

TIMP-1 expression was not affected. Conclusion: Danlou tablet may reduce myocardial necrosis area
and promote infarct healing
prevention and treatment of early left ventricular remodeling by inhibiting+ MMP-2
MMP-9 protein expression;Danlou tablet has no significant effect on the expression of TIMP-1
suggesting the inhibition of MMP activity may not be achieved by TIMP.
