目的: 研究马齿苋不同提取物对D-半乳糖致衰老小鼠空间学习能力的影响
并探讨其作用机制。 方法: 小鼠随机分为正常组、模型组、马齿苋正丁醇提取物(0.5
0.25 g ·kg-1)组和马齿苋乙醇提取物(1
0.5 g ·kg-1)组。除正常组颈背部SC生理盐水外
其他各组均sc D-半乳糖(1 g ·kg-1)连续给药和造模42 d。用Morris水迷宫测定衰老小鼠空间学习记忆能力。同时测定小鼠脑组织丙二醛(MDA)含量、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活性。 结果: 马齿苋正丁醇提取物(0.5
0.25 g ·kg-1 )组和马齿苋乙醇提取物(1 g ·kg-1)组均可使衰老小鼠逃避潜伏期明显缩短;马齿苋正丁醇提取物(0.5 g ·kg-1)组和马齿苋乙醇提取物(1 g ·kg-1)组均可使衰老小鼠1 min站台穿越次数明显增多;马齿苋正丁醇提取物(0.5
0.25 g ·kg-1)组可降低脑组织MDA含量
提高SOD和GSH-Px活性;马齿苋乙醇提取物(1 g ·kg-1)组可降低脑组织MDA含量。 结论: 马齿苋不同提取物均能明显改善衰老小鼠空间学习记忆能力
其机制可能与提高SOD
GSH-Px活性
降低MDA含量有关。马齿苋正丁醇提取物好于马齿苋乙醇提取物。
Objective: To study effects of extracts from Portulacae Herba on spatial learning performance in senile mice induced by D-gal
and to explore the related mechanism. Method: The experimental mice were randomly divided into 5 groups:control group
model group and the n-butanol extracts from Portulacae Herba (0.5
0.25 g ·kg-1 ) groups
the ethanol extracts from Portulacae Herba (1
0.5 g ·kg-1) groups.Except the control group was given sc normal saline in the cervical back part.The rest of the mice were given sc D-gal(1 g ·kg-1) once daily continually for 42 days
while the mice were given medicines accordingly.Learning and memory performances were tested with Morris water maze test
and the content of malondialdehyde (MDA) in brain tissue of the senile mice
and the activities of superoxide dismutase(SOD) and lutathione peroxidase(GSH-Px)were determined. Result: The n-butanol extracts from Portulacae Herba (0.5
0.25 g ·kg-1) groups and the ethanol extracts from Portulacae Herba (1 g ·kg-1) group could obviously shorten the escape latency in senile mice.The n-butanol extract (0.5 g ·kg-1)and the ethanol extracts from Portulacae Herba (1
0.5 g ·kg-1) groups showedincrease in the frequency of passing the plateform within 1 min for the mice.The ethanol extracts (1 g ·kg-1) groups could decreases content of MDA and improve activity of SOD and GSH-Px in brain tissue.The ethanol extracts (1 g ·kg-1) groups could decrease content of MDA. Conclusion: The different extract from Portulacae Herba can ameliorate the spatial learning and memory performances of senile mice.The mechanism may be related to decrease in MDA in brain tissue and increase in activities of SOD and GSH-Px.The n-butanol extract from Portulacae Herba appears to be better than the ethanol extract from Portulacae Herba.
