ObJective: The effects of the different Salvianolic acid B (Sal B) and Paeonol (Pae) drug combinations on isolated vascular and acute myocardial ischemia-reperfusion inJury (MI/RI) were investigated in rats. Method: A increase-decrease design method of experiment with baseline geometric proportion was established to design Salvianolic acid B and Paeonol drug combinations. The changes in isometric tension were recorded by force transducers connected to the system. Vaso-relaxant effects of the drug combinations were observed on the contraction of rat thoracic aorta rings. Acute myocardial ischemia model was made by ligation of left anterior descending artery in the anesthetized open chest rats.Changes in baseline geometric was designed for the different combinations
the effects of different combinations on electrocardiogram(ECG)
myocardial infarction square(MIS)
malondialdehyde(MDA)
superoxide dismutase(SOD)and nitrogen monoxidum(NO) were measured
and effects on isoenzyme of creatine-kinase(CK-MB) were also determined. Result : The drug combinations on vasorelaxant activity was superior to model group and can reduce the degree of myocardial ischemia(∑-ST) and the infarction square(MIS)
Moreover
it also can reduce MDA and CK-MB in serum
and improve SOD and NO activity in MI/RI. The ratio of 3 ∶1 was the best in the drug combinations. Conclusion :The drug combinations of Sal B and Pae have some protective effects on acute myocardial ischemia-reperfusion inJury in rats. Some synergistic or additive effects may exist between Sal B and Pae.
