用Bliss法分别计算半数致死量(LD50)及其95%可信度。 结果 :小鼠口服山豆根乙醇热回流液和渗漉液的LD50和95%可信限分别是27.135(24.869~29.622 g ·kg-1)和13.399(12.016~14.899 g ·kg-1)
分别相当于人日用量的316.6倍和156.3倍
主要毒性症状谱为多动、烦躁、间歇性抽搐、呼吸急促等
最后因呼吸衰竭而死亡
成活动物体重与空白对照组相比有差异
药后7 d差异消失。 结论 :山豆根乙醇渗漉液的毒性大于热回流液
说明乙醇提取温度和浓度是影响山豆根急性毒性结果表达的关键因素
尤以温度为显著。
Abstract
ObJective: To compare the influence of different alcohol extraction on the acute toxicity of Sophorae Tonkinensis Radix et Rhizome
caused by different extracted crafts. Method : Separately using percolate and reflux extracted methods to prepare the extracts of different processed products. The classical acute toxicity methods were used in this study

death time and accumulated death rate were recorded and the LD50were calculated by Bliss method. Result : The LD50 and its 95% confident limit of reflux and percolation extracts on mice were respectively 27.135(24.869-29.622)g ·kg-1 and 13.399(12.016-14.899)g ·kg-1
which are equal to 316.6 and 156.3 times of the daily dosage in clinical
the main acute toxical symptoms such as: hyperactivity
restlessness
intermittent convulsion
breathlessness and died of respiratory failure eventually. The body weight between survival and normal mice had obvious difference (P<0.05)
this difference disappeared after 7 d administration. Conclusion : The acute toxicity of different alcohol extraction of Sophorae Tonkinensis Radix et Rhizome is: percolation extracts>reflux extracts
explaining the key factors are temperature and concentration
especially the temperature
that affect the acute toxicity result expression of different alcohol extraction.
