目的 :探讨熊果酸和槲皮苷的镇静催眠作用是否与睡眠的一氧化氮调节和免疫调节机制两个方面有关。 方法 :熊果酸、槲皮苷注射正常大鼠和睡眠剥夺大鼠后
进行指标测量。NO含量测定采用Griess法
白细胞介素-1β(IL-1β) 和肿瘤坏死因子-α(TNF-α)采用放免法测定。 结果 :①正常大鼠腹腔注射熊果酸2 h后
各脑区内NO水平升高
IL-1β水平升高
TNF-α水平也略有升高
但与空白组比较均没有显著性意义。②正常大鼠腹腔注射槲皮苷2 h后
各脑区中NO水平下降
IL-1β和TNF-α水平均有升高趋势
其中下丘脑内TNF-α含量增加显著
与空白组比较有显著性意义。③睡眠剥夺大鼠腹腔注射熊果酸4 h后
脑内NO水平显著升高
与空白组和模型组比较均有显著性意义;血清中IL-1β含量增加
与空白组和模型组比较均有显著性意义。④睡眠剥夺大鼠腹腔注射槲皮苷4 h后
脑内NO含量显著升高
与空白组和模型组比较均有显著性意义;IL-1β含量显著增加
与空白组比较有显著性意义;下丘脑中的TNF-α含量升高
与空白组比较具有显著性意义。 结论 :熊果酸对正常大鼠和睡眠剥夺大鼠睡眠的影响可能与睡眠调节的一氧化氮和免疫调节机制有关;槲皮苷对睡眠剥夺大鼠睡眠的影响可能与一氧化氮和免疫调节机制有关
对正常大鼠
免疫调节机制也可能参与其中。
Objective: This experiment aim to observe the relationship between sedative and hypnic actions of the Ursolic acid and quercitrin with nitric oxide (NO) and immune regulation mechanism of sleep. Method: The experiment respectively surveyed the indicatrix after intraperitoneal injection of ursolic acid and quercitrin in brain of both nomal and sleep-deprived rats. The assaying of NO is through the method of Griess. The assaying of LI-α and TNF-β is through the method of radioimmunity. Result: ① Two hours after intraperitoneal injection of ursolic acid in normal rat
the level of NO in every encephalic region and IL-1β in blood serum
hippocampus and hypothalamus were enhanced
as well as the amount of TNF-α was slightly increased. But compared with blank group
all results wasn’t different significant. ②Two hours after intraperitoneal injection of quercitrin in normal rat
the amount of NO in every encephalic region was cut down;The levels of LI-1β and TNF-α in blood serum and each encephalic was raised
especially the amount of LI-1β in hypothalamus was increade markedly compared with blank group. ③ Four hours after intraperitoneal injection of ursolic acid in sleep-deprivated rat
the amount of NO in brain were increased compared with either blank or model group
having significant different;the amount of IL-1β in blood serum was significantly elevated compared with either blank or model group. The levels of LI-1β and TNF-α in blood serum
hippocampus and hypothalamus were enhanced
but it was insignificant. ④ Four hours after intraperitoneal injection of quercitrin in sleep-deprived rats
the level of NO in every encephalic region were enhanced
compared with blank and model group
it had significant difference. The level of IL-1β in blood serum was enhanced compared with blank and model group
it had significant different. The amount of LI-1β in hippocampus and hypothalamus was increased significantly compared with blank group. The level of TNF-α in hypothalamus was enhanced significantly. Conclusion: The sedative and hypnic actions of ursolic acid and quercitrin are related with NO and immune regulation mechanism of sleep. Quercitrin of influencing sleep-deprived rat’ sleep is probably related to NO and immunoregulation mechanism. For nomal rat immunoregulation mechanism may be participate in.
