Objective: To observe the effect of curcumin on anti-liver fibrosis and its mechanism. Methods: Model of liver fibrosis was induced by intraperitoneal injection of dimethylnitrosamine. Rats were divided into 5 groups after four weeks
each group of rats were treated with corresponding drug. The serum HA
PCIII and Hyp in liver tissue were determined. The changes of liver and expression of PDGF-BB
PDGFRβ
ERK1 of liver were observed by immunohistochemistry. The level and of PDGFRβ
ERK1 mRNA were observed by RT-PCR. Results: Curcumin could obviously decrease relevant index on liver fibrosis and the level of Hyp. Pathological results indicated curcumin could obviously improve the pathological lesion of rat liver fibrosis. The protein expression of PDGF-BB
PDGFRβ
ERK1 was inhibited
and the mRNA expression of PDGFRβ
ERK1 mRNA was down regulated by curcumin. Conclusion: Curcumin is effective in treatment of liver fibrosis
and its mechanism of anti-liver fibrosis is related to inhibiting the expression of PDGFRβ
