Objective: To study the effects of Xiaojia Decoction on expression of Vascular Endothelial Growth Factor in transplantation tumors of the breast cancer cell line MDA-MB-435 in nude mice. Methods: The MDA-MB435 human breast cancer cells cultured in vitro were subcutaneously inoculated into BALB/c nude mice to establish the transplanted tumor model. The mice were then randomized into five groups and treated with Xiaojia Decoction or other remedies for 6 weeks. Then we sacrificed the mice and weighed the tumor
calculated the inhibitory rate of tumor weight
the expression of VEGF on mRNA and protein level was determined by means of RT-PCR and immunohistochemical assay. Results: Xiaojia Decoction could inhibit the growth of tumor conspicuously
lower the expression of both mRNA and protein level of VEGF(P<0.05)
especially for the medium-dose group. Conclusion: This Decoction's effects on down-regulating VEGF have close relation with its function of inhibiting the tumor growth.
