摘要

目的:建立五种不同的氨基酸的HPLC-ELSD分析方法

并对其响应因子进行分析。方法:KromasilC18色谱柱(4.6×250mm

5μm)为固定相

柱温30℃;流动相为含0.1%三氟醋酸的水

流速0.3mL·min-1;ELSD检测参数为:漂移管温度105℃

气体流速2.60L·min-1。通过测定不同氨基酸组分的线性方程

考察其响应因子。结果:不同氨基酸线性方程的斜率具有显著的差异。结论:不同氨基酸HPLC-ELSD检测的响应因子具有明显的不同。

Abstract

Objectives:To develop the HPLC-ELSD method to detect the different amino acids and analyze the response factors of different constituents detected by ELSD.Methods:The stationary phase was Kromasil C18 column(4.6×250mm

5μm)at 30℃.The mobile phase was water contained 0.1%TFA with flow rate was 0.3mL·min.The chromatogram was recorded by ELSD with drift tube temperature was 105℃ and gas flow rate was 2.60L·min-1.The response factors were analyzed by compare the standard curves of different amino acids.Result:The difference between the slops of standard curves was found.Conclusion:The response factor of different amino acids detected by ELSD was greatly different.
