Objective: To optimize extraction process of total valepotriates from Valeriana jatamansi by Plackett-Burman combined with central composite design-response surface methodology. Method: Based on single factor test
Plackett-Burman test was adopted to screen main influencing factors;Extraction technology of total valepotriates was optimized by central composite design-response surface method
with the concentration of ethanol
soaking time
ultrasound time as independent variables
dependent variable was the content of total valepotriates
through complete quadratic response surface regression fitting of independent variables and the dependent variable
optimum technology was determined by intuitive analysis of 3D surface chart
and predictive analysis was used. Result: Optimum extraction technology was as following:the concentration of ethanol 65%
soaking time 3 hours
liquid-solid 10: 1
ultrasonic extracted 2 times at room temperature
30 minutes per time. Conclusion: This optimized process was simple
