Objective: To optimize extraction and purification technology of salidroside from Ligustrum lucidum. Method: The content of salidroside was determined by HPLC
with composite score of the content of salidroside and yield of extract as index
orthogonal test was adopted to optimize extraction technology with solvent
liquid-solid ratio
extraction time and times as factors;model of macroporous resin was screened by single factor test
and its purification technology was optimized. Result: Optimum extraction technology was as following:reflux extracted 3 times with 6 times the amount of water
each time for 1 hour;optimized purification technology of D4020 type resin was:the concentration of sample solution 0.5 g·mL-1
adsorption velocity for 1 BV·h-1
eluted with5 BV 20% ethanol
collected eluate
vacuum recovery
then received salidroside crude product with purity of 16.34%. Conclusion: By following these optimized extraction and purification conditions
the purity of salidroside in L. lucidum could increase 2.12 times.
