Objective: To optimize extraction process of total flavonoids from Callicarpa macrophylla (TFCM) and estimate its anti-inflammatory
antalgic and hemostatic activities. Method: With yield of total flavonoids as index
on the basis of single factor test
effect of the concentration of ethanol
liquid-solid ratio
extraction times and time on extraction technology of TFCM;Anti-inflammatory
antalgic and hemostatic activities of TFCM were evaluated by xylene-induced ear edema
acetic acid-induced writhing
glass capillary tube method in mice
respectively. Result: Optimal extraction parameters of TFCM were ethanol concentration 90%
ratio of solid to liquid 1:20
extraction temperature 80℃
extracted 2 times with 2 h per time.Under these conditions
yield of TFCM was 63.1 g·kg-1.After purified by macroporous resin and polyamide resin
purity of TFCM was up to 53.3%.TFCM displayed potent anti-inflammatory
antinociceptive activity and significantly shortened the blood coagulation time in mice at the dose of 200 and 400 mg·kg-1
but TFCM merely exhibited antalgic activity at the dose of 100 mg·kg-1. Conclusion: Optimized process could achieve purpose of enrichment of active ingredients
TFCM possessed significant anti-inflammatory
antalgic and hemostatic activities with dose-related.
