目的: 观察抗放射性肺炎方(anti-radiation pneumonia decoction
ARPD)水提醇沉精制液和水煎剂治疗放射性肺损伤的等效性。 方法: 采用水提醇沉法初步精制ARPD水煎剂。将60只大鼠随机分为模型组、水煎剂组、精制液组和正常对照组。前3组均行直线加速器6MVX射线17Gy全胸单次照射。照射后每日分别给予ig生理盐水、ARPD水煎剂和精制液各35 g·kg-1
1次/d
每次10 mL·kg-1。各组大鼠分别于照射后第7

检测肺组织和血清中超氧化物歧化酶(SOD)、丙二醛(MDA)和羟脯氨酸(HYP)含量。 结果: ①模型组大鼠肺组织病理检查发现炎症改变及胶原增生
肺组织和血清SOD活力下降
而 MDA和HYP升高
与同期对照组比有显著差异(P<0.01);②ARPD水煎剂组、精制液组两组肺组织病理检查炎症改变及胶原增生明显轻于同期模型组
两中药间无明显差异;③SOD活力下降
MDA和HYP升高
模型组甚于水煎剂组和精制液组(P<0.01)
两中药制剂间无显著差异。 结论: ①早期应用ARPD可以明显减轻肺的放射性炎症反应
降低肺纤维化的程度;②ARPD精制液和水煎剂的抗放射性肺损伤疗效相当;③水提醇沉法是ARPD纯化精制的可行的工艺方法。
Objective: Equivalence studys of refined extract and crude solution of anti-radiation pneumonia decoction (ARPD) on radiation-induced lung injury. Method: Refined extract of ARPD was made by alcohol purification from crude solution. Experiment group: model group
crude solution treatment group
refined extract treatment group and control group. Rats in model group
crude solution treatment group and refined extract treatment group were thoracically exposed to 6 MVX-rays at a dose of 17 Gy. The rats were treated with different solutions by gastrogavage once a day just after X-ray exposure. Rats in crude solution treatment group and refined extract treatment group were treated with 3.5 g·mL-1 crude and refined ARPD respectively
and rats in model group and control group were gavaged with 0.9% sodium chloride
each time 10 mL·kg-1. The rats were sacrificed at the day of 7

45 and 60 after X-ray exposure. The levels of superoxide dismutase(SOD)
malondialdehyde (MDA) and hydroxyproline (HYP) in lung tissues and sera were measured simultaneously. Result: ①Pathological examination of lung tissue of model group revealed inflammatory changes and proliferation of collagenous fibers
in lung tissue and serum SOD activity decreased
while the content of HYP and MDA increased
much higher than the concurrent control group (P<0.01)
with a statistically significant difference; ②The inflammatory reactions and collagenous proliferation in crude solution treatment group and refined extract treatment group were much slighter
compared with the same period of model group
but there was no difference between these two treatment groups;③The reduction of SOD activity and the rises of content of MDA and HYP in the model group severely greater than the crude solution treatment group and refined extract treatment group(P<0.01)
while no significant difference between two treatment groups. Conclusion: ①ARPD may reduce the inflammatory reaction of radiation-induced lung injury and reduce pulmonary fibrosis caused by radiation injury. ②There are similar anti-radiation effects on treating lung injury between refined extract and crude solution of ARPD.③The method of water extraction and alcohol precipitation is viable in purifying ARPD.
