目的: 探讨蒿鳖养阴软坚方对CCl4复合因素所致大鼠肝纤维化的预防作用及可能的作用机制。 方法: 将大鼠随机分为正常对照组
模型组
蒿鳖养阴软坚方高、中、低剂量组(8.20

0.82 g·kg-1)
复方鳖甲软肝片组(0.54 g·kg-1)
秋水仙碱组(1×10-4 g·kg-1)。第1次sc CCl4 5 mL·kg-1
从第2次开始每隔3 d sc 40%CCl4花生油3 mL·kg-1至第12次; 模型组和治疗组隔日ig 30%乙醇(1 mL/只)
各治疗组每日给药1次
正常组和模型组均给予相应体积的蒸馏水
每周6 d(停药1 d)
共6周。观测肝组织形态和胶原蛋白
测定血清肝功能丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST);放免法测定血清透明质酸(HA)、IV型胶原(IV-C)、III型前胶原(PCIII)
层黏连蛋白(LN)水平;紫外-可见分光光度法测定血清丙二醛(MDA)水平;检测肝组织中羟脯氨酸
胶原蛋白及白蛋白含量。 结果: 模型组大鼠肝组织内肝细胞广泛变性及坏死
大量炎性细胞浸润
类假小叶形成
大量胶原纤维形成;蒿鳖养阴软坚方高剂量组的肝脏病理变化程度较模型组明显减轻且无类假小叶形成;模型组的ALT
AST
HA及PCIII含量均高于正常对照组( P<0.01
P<0.05);各蒿鳖养阴软坚方给药组ALT
AST
HA及PCIII高剂量组显著低于模型组(P<0.05); MDA及胶原蛋白的含量均低于模型组(P<0.01)
各蒿鳖养阴软坚方给药组白蛋白均高于模型组(P<0.01)。 结论: 蒿鳖养阴软坚方对四氯化碳复合因素诱导的大鼠肝纤维化有预防作用;其作用机制可能与抗脂质过氧化损伤
抗炎
抑制胶原蛋白的合成等有关。
Objective: To study the preventive effects of Haobie Yangyin Ruajian prescription (HBYYRJ) on rat hepatic fibrosis induced in rats by CCl4 and explore the effective mechanisms. Method: Rats were randomly divided into 7 groups:normal control group

40% CCl4-peanut oil 3 mL·kg-1 was given by sc every three days from the second time to the twelfth time; 30% alcohol (1 mL) was ig given to model group and therapeutic groups every other day
drugs were given to the rats in therapeutic groups for six consecutive weeks
distilled water was given to the rats of normal and model groups. Histological observation of the liver tissue and collagen content were carried out. Blood serum was used for measuring the contents of alanine transaminase(ALT)
aspartate aminotransferase(AST)
hyaluronic acid(HA)
laminin(LN)
type III procollagen(PCIII)
type IV collagen(IVC)
albumin(ALB). The content of malondialdehyde (MDA) was detected by ultraviolet visible spectrophotometry. The content of hydroxyproline

Conclusion: HBYYRJ has preventive effects on hepatic fibrosis in rats induced by CCl4.The mechanism might be associated with anti-lipid peroxidation
antiinflammation and restraining the compose of collagen protein.
