目的: 观察茵黄合剂对雌孕激素共诱导妊娠肝内胆汁淤积症(intrahepatic cholestasis of pregnancy
ICP)大鼠胆汁酸转运蛋白多药耐药相关蛋白2(multidrug resistance-associated protein 2
MRP2)与胆盐输出泵(bile salt export pump
BSEP)表达的影响
探讨茵黄合剂治疗妊娠肝内胆汁淤积症可能的分子机制。方法: 运用雌孕激素联合的方法建立孕鼠妊娠肝内胆汁淤积症模型
随机分为正常组、模型组、茵黄合剂低剂量组(9 g·kg-1·d-1
ig)和茵黄合剂高剂量组(18 g·kg-1·d-1
ig)
分别给予生理盐水、低剂量与高剂量的茵黄合剂5 d
取血
检测血清总胆汁酸(total bile acid
TBA)、总胆红素(total bilirubin
TBil)、丙氨酸转氨酶(alanine transarninase
ALT)、天冬氨酸转氨酶(aspartate aminotransferase
AST)活性
留取肝脏组织
置于液氮中保存
分别运用实时定量PCR法及蛋白免疫印迹法检测胆汁酸转运蛋白MRP2 BSEP mRNA与蛋白表达情况。结果: 与正常组比较
模型组大鼠血清TBA
TBil水平增加
MRP2
BSEP mRNA与蛋白表达水平降低(P<0.01)
模型组ALT
AST含量与正常组比较虽有增加
但差异无统计学意义。与模型组比较
茵黄合剂低、高剂量均能降低妊娠肝内胆汁淤积症模型大鼠血清TBA
TBil水平(P<0.01)
并且可明显增加肝脏组织MRP2
BSEPmRNA与蛋白表达水平
P<0.01。结论: 茵黄合剂治疗妊娠肝内胆汁淤积症的分子机制可能是通过增加肝脏组织胆汁酸转运蛋白MRP2
BSEP
mRNA与蛋白表达水平实现的。
Objective: To investigate the effect of Yinhuang mixture on the expression of multidrug resistance-associated protein 2 (MRP2) and bile salt export pump (BSEP) in rats with intrahepatic cholestasis of pregnancy induced by estrogen and progesterone. Method: The animal model of intrahepatic cholestasis of pregnancy was induced by 17-α-ethinylestradiol and progesterone and these rats were randomly divided into four groups
including normal group
model group
low dosage and high dosage of Yinhuang mixture groups
either saline or low and high dosage of Yinhuang mixture were given. The serum levels of total bile acid (TBA) total bilirubin (TBil) alanine transarninase(ALT) aspartate aminotransferase
(AST)were examined
and the expression of MRP2 and BSEP were detected by real-time RT-PCR and Western blotting analysis. Result: Compared with the model group
the serum levels of TBA and TBil were remarkably decreased in low dosage and high dosage of Yinhuang mixture groups
in addition
the expression of MRP2 and BSEP were significantly increased in these groups. Conclusion: The therapeutic effects of Yinhuang mixture on intrahepatic cholestasis of pregnancy may upregulate the expression of MRP2 and BSEP.
