目的: 研究四妙勇安汤含药血清对高糖/高胰岛素诱导的兔血管平滑肌细胞(VSMC)增殖的影响及其作用机制。方法: 9只新西兰大耳白兔随机分为3组
每组3只
分别给予蒸馏水10 mL/次
2次/日;辛伐他汀5 mg·kg-1
1次/日;四妙勇 安汤按生药量11.25 g·kg-1
2次/日
连续3 d
血清药理学方法制备含药血清
高糖/高胰岛素(4 500 mg·L-1葡萄糖+100 U·L-1 胰岛素)诱导兔VSMC增殖
实验分为空白组(10%FBS的正常DMEM)、正常组(10%FBS的正常DMEM+5%正常血清)、模型组(10%FBS的高糖DMEM+100 U·L-1胰岛素+5%正常血清)、西药组(10%FBS的高糖DMEM+100 U·L-1胰岛素+5%辛伐他汀血清)、中药组(10%FBS的高糖DMEM+100 U·L-1胰岛素+5%四妙勇安汤血清)
四唑盐比色(MTT)法检测细胞增殖
酶联免疫吸附测定(ELISA)法检测细胞培养上清液中白介素-6(IL-6)和肿瘤坏死因子(TNF-α)的含量。结果: 高糖/高胰岛素可诱导VSMC增殖
与正常组比较模型组MTT的吸光度(A)、细胞培养上清液IL-6
TNF-α的含量显著升高(P<0.05);与模型组比较
中药组、西药组均能降低MTT的A、细胞培养上清液IL-6和TNF-α的含量(P<0.05)。结论: 四妙勇安汤对高糖高胰岛素诱导的平滑肌细胞增殖有一定的抑制作用
其机制之一可能与减少IL-6和TNF-α分泌有关。
Objective: To observe the anti-proliferative effects and possible mechanisms of serum containing Simiao Yongan decoction(SYD) on rabbit aortic smooth muscle cells(VSMC) during the insulin resistance state. Method: Nine New Zealand big-eared rabbits were randomly divided into 3 groups with 3 rabbits each group.Each groups were separately given distilled water 10 mL
simvastatin 5 mg·kg-1 and SYD at dosage of 11.25 g·kg-1 each time and twice a day for 3 consecutive days.Serum containing SYD was prepared by serum pharmacological method
the proliferation of VSMC was induced by high concentrations of glucose and insulin(4 500 mg·L-1 glucose+100 U·L-1insulin).The VSMCs were divided into following groups:blank group (10%FBS normal glucose DMEM)
normal group (10%FBS normal glucose DMEM+normal serum containing)
model group (10%FBS glucose DMEM+100 U·L-1 insulin+normal serum containing)
western medicine group (10%FBS high glucose DMEM+100 U·L-1 insulin+serum containing simvastatin)
traditional Chinese medicine group (10%FBS high glucose DMEM+100 U·L-1 insulin+serum containing SYD).Cell proliferation was measured by methyl thiazolyl tetrazolium (MTT) assay
content of IL-6 and TNF-α in cell culture supernatant was detected by enzyme linked immunosorbent assay (ELISA) method. Result: The proliferation of VSMC was markedly induced by high concentrations of glucose and insulin.Compared with the normal group

Chinese medicine group and western medicine group

the content of IL-6 and TNF-α of cell culture supernatant was reduced(P<0.05). Conclusion: Serum containing SYD can inhibit proliferation of the smooth muscle cells induced by high glucose and insulin
one of the possible mechanisms may be related to reducing secretion of IL-6 and TNF-α in smooth muscle cells.
