Objective: To optimize ultrasonic-assisted enzymatic hydrolysis technology of baohuosideⅠ. Method: The content of baohuosideⅠ was determined by HPLC
chromatographic conditions were as follows:Diamond C18 column(4.60 mm×250 mm
5 μm)
mobile phase of acetonitrile-water(25:75)
flow rate 1.0 mL·min-1
column temperature 30℃
detection wavelength 270 nm.Taking bioconversion rate of baohuosideⅠ as index
based on single factor tests

temperature
enzyme dosage and reaction time on preparation technology were investigated L9(34) orthogonal design. Result: Optimal enzymatic conditions were:ultrasonic power of 200 W
pH of 5.0
reaction temperature at 40℃
reaction time of 4 h
enzyme dosage 5%.Under these conditions
conversion rate of baohuosideⅠ was 97.4%. Conclusion: Ultrasonic-assisted enzymatic method could improve conversion rate of baohuosideⅠ with low reaction temperature
