目的: 观察干姜、肉桂、丁香、吴茱萸、小茴香、高良姜、荜茇、荜澄茄、花椒、胡椒10味温中散寒药对胃实寒证大鼠胃黏膜组织病理学的影响,以期观察温中散寒药的温胃、护胃作用,探讨温中散寒药的作用机制与药效本质。 方法: SD雄性大鼠分为15个组:空白组、模型组、附子组、干姜组、肉桂组、丁香组、吴茱萸组、小茴香组、高良姜组、荜茇组、荜澄茄组、花椒组、胡椒组、石膏组、黄连组。灌服知母水提液17.5 g·kg-1,2次/d,连续2 d建立大鼠胃实寒证模型。附子组13.5 g·kg-1,干姜、胡椒组8.1 g·kg-1,肉桂、吴茱萸组4.1 g·kg-1,丁香、荜茇、荜澄茄组2.7 g·kg-1,小茴香、高良姜、花椒组5.4 g·kg-1,石膏组54 g·kg-1,黄连组4.5 g·kg-1,2次/d,连续3次。肉眼观察胃黏膜变化,光镜下观察胃黏膜组织病理学改变并根据胃组织病理改变的评分标准进行评分。 结果: 肉眼观察,与空白组比较,模型组胃黏膜表面颜色灰暗、皱缩、无光泽,有大小不等的出血点;与模型组比较,10个温中散寒药组的胃黏膜均有所改善。光镜下观察,与模型组比较,除高良姜组外其余9个组均能明显减少炎细胞浸润(P<0.05或P<0.01);10个温中散寒药组均能明显减少胃黏膜腺体的破坏(P<0.01)。 结论: 10味温中散寒药通过改善大鼠胃黏膜的破坏从而发挥其治疗胃实寒证的药效。

Gypsum Fibrosum group and Rhizoma Coptidis group.the stomach cold syndrome model in rats was established by water extract of Rhizoma anemarrhenae 17.5 g·kg-1
twice per day
lasting two days. the dosage of Radix Aconiti Lateralis Preparatax group was 13.5 g·kg-1
the dosage of Rhizoma Zingiberis and Fructus Piperis Nigri group was 8.1 g·kg-1
the dosage of Cortex Cinnamomi and Fructus Evodiae group was 4.1 g·kg-1
the dosage of Fructus Caryophylli and Fructus Piperis Longi
Piper cubeba group was 2.7 g·kg-1
the dosage of Fructus Foeniculi
Rhizoma Alpiniae Officinarum and Pericarpium Zanthoxyli group was 5.4 g·kg-1
the dosage of Gypsum Fibrosum group was 54 g·kg-1
the dosage of Rhizoma Coptidis group was 4.5 g·kg-1
twice per day in 3 times. The changes of the gastric mucosa were observed by naked eyes and th echanges of pathological section were observed by light microscope
and graded according score criterion of the pathologic tissues of gastric mucosa. Result: Compared with blank group
the stomach mucous membrane in model group had dark color
crimple
hemorrhagic spots and no gloss
compared with model groupthe stomach mucous membrane in the 10 treatment groups was improved.All the treatment groups could significantly decrease the imflammatory cells infiltration(P<0.05 or P<0.01)except the group of Alpinia group and reduce the glands(P<0.01). Conclusion: The ten herbs with warming middle-jiao and dispelling cold have good improvement on the pathologic tissues of gastric mucosa so as to treat stomach cold syndrome.
