Objective: To establish a method for determining the content of berberine in Chaishao Changning capsules and investigate effects of different decoction ways on the content of berberine. Method: HPLC was adopted to determine the content of berberine with DIKMA ODS-C18 column (4.6 mm×250 mm
5 μm)
mobile phase of acetonitrile-0.1% phosphoric acid(per 100 mL plus sodium dodecyl sulfate 0.1 g)(50:50) and detection wavelength at 345 nm.Effects of mixed decoction and single decoction of Coptidis Rhizoma with other herbs on the content of berberine were investigated. Result: Berberine had a goog linear relationship with 2.4-24 mg·L-1 with r=1
the average recovery was 101.14% with RSD of 2.48%.Average contents of berberine from decoction and separate decoction samples were 0.128 2
0.078 6 mg·g-1 with RSD of 1.02%
1.25%
respectively. Conclusion: The content of berberine from mixed decoction samples was significantly higher than single decoction samples
but it should be made conclusion through clinical trials.
