目的:观察壮骨1号方对骨性关节炎的抗损伤作用。方法:选SD大鼠,以泊洛沙姆载药体加载木瓜蛋白酶制成温敏型原位凝胶控释剂,按照0.05 mL(1.6 U)/关节剂量,双侧膝关节注射,50 ℃环境下作用2 h复制大鼠骨性关节炎模型。实验分为正常对照、模型对照组(等容蒸馏水)、壮骨关节丸(1.10 g·kg-1)组与壮骨1号方(2.20,1.10,0.55 g·kg-1)治疗组,氨基葡糖(0.03 g·kg-1)阳性药对照组,ig每天1次,连续治疗8周。用酶联免疫吸附(ELISA)法测定关节液中基质金属蛋白酶-3(MMP-3)及其组织金属蛋白酶抑制因子-1(TIMP-1)含量,计算MMP-3/TIMP-1;观察关节软骨病理组织学变化并进行图像分析。结果:治疗8周后,模型组大鼠关节液中MMP-3/TIMP-1为0.092±0.010,与正常组0.062±0.011差异有统计学意义(P<0.05),用壮骨1号方2.20,1.10,0.55 g·kg-1剂量治疗8周后,MMP-3/TIMP-1明显降低,分别为0.060±0.016,0.060±0.028,0.057±0.027,与模型组比差异有统计学意义(P<0.05或P<0.01)。病理图像分析显示模型组大鼠软骨坏死面积为(37 734±11 504)μm2,与正常组(0.00±0.00)μm2比较差异有统计学意义(P<0.01),壮骨1号方2.20,1.10 g·kg-1治疗8周后,关节软骨坏死面积为(26 601±6 506),(27 061±10 425)μm2,与模型组比较差异有统计学意义(P<0.05)。结论:壮骨1号方具有一定的抗软骨损伤作用,能延缓骨性关节炎的病程进展。
Objective: To explore the anti-injury effect of Zhuanggu Yihao Fang on osteoarthritis (OA). Method: The OA rat model was induced by sustained releasing agent (4% Papain and poloxamer).0.05 mL of the sustained releasing agent (4% Papain and poloxamer) was injected into both knee joint compartments of male SD rats to injury cartilage for a period of 2 h at 50 ℃. 0.55 g·kg-1) groups. The rats with osteoarthritis received daily oral treatments of drugs for 8 weeks. The normal group and the model group received daily oral treatments of isovolumic distilled water for 8 weeks. The contents of matrix metalloproteinases-3(MMP-3) and tissue inhibitar of metalloproteinases-1(TIMP-1) in synovial fluid were detected by enzyme-linked immunosorbent assay (ELISA). The changes in cartilages were evaluated by histopathology and morphometry. Result: MMP-3/TIMP-1 level in normal group was 0.062±0.011
