Objective: To optimize ultrasonic extraction process of triptolide from Tripterygium wilfordii. Method: With yield of triptolide as index
response surface methodology was adopted to optimize extraction technology by taking material-liquid ratio
ethanol concentration
extraction time and ultrasonic temperature as factors.HPLC was adopted to determine the content of triptolide with mobile phase of acetonitrile-water(33:67) and detection wavelength at 218 nm. Result: Optimal extraction conditions were as follows:solid-liquid ratio of 1:12
ethanol concentration 70%
extraction temperature at 35 ℃
extraction time 30 min;under these conditions
yield of triptolide was 0.037 9 mg·g-1
which was close to the predicted value(0.038 6 mg·g-1). Conclusion: This optimized extraction process was stable and reliable
which had obvious advantages by comparing with conventional reflux extraction method.
