Objective:To probe the counteractiv emechanism of “aconite counteracts fritillaria”.Methods:We used Solid phase extraction(SPF)to extract and enrich the components contained in the plasma.Chromatographic condition: Methyl alcohol-water-chloroform-triethylamine(70∶30∶2∶0.2)was used as the mobile phase
the flow rate was 1.0mL·min -1 with detection wavelength at 235nm and column temperature at 40℃; detecting the contents of mesaconitine
aconitine and hypaconitine.Result:By using singly or mixed with thunberg fritillary bulb omni-alkaloids
there were obvious differences between these pharmacokinetic parameters of mesaconitine and aconitine: the elimination half-lifes and AUCs of two aconite alkaloids after used with thuberg fritillary bulb omni-alkaloids were longer or higher than those before matching
and the plasma clearances of two aconite alkaloids after used with thunberg fritillary bulb omni-alkaloids were lower than these before matching(P<0.05).There were unsignificanted differences of hypaconitine used singly and used mixed with thunberg fritillary bulb omni-alkaloids(P>0.05).Conclusion:Retention time and affection time of aconite alkaloids after used with thunberg fritillary bulb omni-alkaloids were longer than these before matching.The statement of “aconite counteracts fritillaria” is preliminarily validated by our pharmacokinetic studies.
