目的:观察桂枝芍药知母汤对免疫性关节炎的治疗作用
并初步探讨其作用机制。方法:以弗氏完全佐剂诱导产生免疫性关节炎模型
用不同剂量桂枝芍药知母汤(GSZ汤20.6、10.3g.kg-1)给模型动物灌胃用药
观察用药后动物致炎对侧膝关节滑膜组织病理改变与RANKL表达。结果:桂枝芍药知母汤用药4周后
可明显减轻关节肿胀程度
使关节指数减小(P<0.05orP<0.01);病理镜检显示
大鼠膝关节滑膜细胞和纤维组织增生减轻
浸润细胞数减少;滑膜组织RANKL表达下降(P<0.01)
本实验条件下
GSZ汤20.6g.kg-1剂量时的疗效与MTX相当。结论:桂枝芍药知母汤对免疫性关节炎大鼠的关节损伤有治疗和保护作用
其机制可能与抑制炎症反应并降低RANKL表达有关。
Objective:To observe the therapeutic effect of Guizhi-Shaoyao-Zhimu(GSZ) decoction on imunologic arthritis and investigate the mechanism of this drug on the disease.Methods:Rats were prepared for immunologic arthritis models induced by Freund’s complete adjuvant(FCA).Different doses of GSZ decoction(20.6 g·kg-1
10.3 g·kg-1) were administrated on the rat model.The counter-lateral synovial tissues of the knee joint were taken for observing pathological changes and the expression of RANKL.Results:In a 4-week of administration of GSZ
lessened swelling of the joints and arthritis index was observed(P<0.05 or P<0.01).The pathological findings suggested there was less severe inflammation in synovium of the joints and fibrous tissues and decreased infiltrating cells.The RANKL expression in synovial tissues tended to decrease(P<0.01).Moreover
the findings suggest that GSZ 20.6 g·kg-1 can be favorable with respect to MTX in treatment of immunologic arthritis.Conclusions:GSZ Decoction can protect the joint impairment of immunologic arthritis rats.The therapeutic mechanism maybe is relevant to inhibit inflammation and the expression of RANKL.
