目的:连续观察45 d给大鼠柴胡总皂苷醇洗脱精制样品导致大鼠长期毒性损伤程度及恢复情况。方法:分别给大鼠灌胃高、中、低剂量的柴胡总皂苷醇洗脱精制样品连续45 d
按柴胡总皂苷计算
高、中、低剂量组分别为60
30
15 mg·kg-1
除观察一般状况外
检测血常规
血清肝功、肾功、脂质代谢、糖代谢相关指标;剖杀大鼠
精密称取心、肝、脾、肺、肾脏质量
计算脏体比值;进行肝脏病理组织学检查。剩余大鼠停止给药
恢复30 d
观察大鼠恢复情况
并进行上述相同指标的检测。结果:不同剂量的柴胡总皂苷醇洗脱精制样品在给药后45 d可导致大鼠体重下降
导致血中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、总胆红素(TBI)、总蛋白(TPC)、尿素氮(BUN)水平明显增高
白蛋白(ALB)明显降低
导致肝脏质量和肝体比值增大
病理学检查可见不同程度的肝组织损伤;上述变化随剂量的增加而逐渐加重
与空白组比较有明显差异。对碱性磷酸酶(AKP)、血糖(Glu)、胆固醇(CHO)、肌酐(Cr)等指标和血常规影响不明显。恢复期大鼠上述各指标与空白组比较均无显著性差异。结论:长时间给予一定剂量的柴胡总皂苷醇洗脱精制品可造成对大鼠明显的蓄积毒性
其毒性损伤部位以肝、肾损伤为主
经过30 d恢复期观察
其脏器损伤基本可恢复。
Objective: To observe the chronic toxicity damage of the refined products of bupleurum saikosaponin with alcohol elution to rats who were continuously administrated for 45 days and its recovery condition.Method:Separately lavage different dose refined products of Bupleurum saikosaponin with alcohol elution to rats
and the high
middle and low lose-group are separately 60
30
15 mg·kg-1 caculated by total saikosaponin
the general conditions were observed and the related indexes were detected
such as the routine control of blood
hepatic function
renal function
the metabolism condition of lipids and the glycometabolism indexes;the rats were dissected and precisionly weigh the weight of heart
liver
spleen
lung
renal
and calculated the ratio of organ to body;thehistopathologic examination of hepatic tissue were examined.The remaining rats were stopped administrating
and recovered for 30 days
observed the recovery condition of rats
and the above indexes were detected.Result:Different dose refined products of Bupleurum saikosaponin with alcohol elution could induce the rats’ body weight decreasing
the level of alanine aminotransterase(ALT)
aspartate aminotransterase(AST)
total bilirubin(TBI)
total protein(TPC)
and blood urea nitrogen(BUN) in serum obviously increasing
alburnin(ALB) in serum obviously decreasing
the liver weight and the ratio of liver to body increasing
and the hepatic tissue damage was obvious in the histopathologic examination of hepatic tissue;the above-mentioned change gradually aggravated with dose increasing
and it was the obvious discrepancy compared with control group.The influence on the content of alkaline phosphatase(AKP)
glucose(Glu)
cholesterol(CHO) and Cr was not obvious.The above indexes of recovered rats had no obvious discrepancy compared with control group.Conclusion:The long term and certain dose administration of refined products of Bupleurum saikosaponin with alcohol elution to rats could induce obvious cumulative toxicity
and the mainly toxical injury sites were liver and renal
after the 30 d recovery period observation
the toxicity was basically recovered.
