摘要

目的:建立适合于瓜蒌种子的蛋白质提取方法。方法:比较水溶液直接提取法（3种）、三氯乙酸（TCA）-丙酮法、三羟甲基氨基甲烷（Tris）-丙酮法、蛋白质提取液提取法等6种蛋白质提取方法所获瓜蒌种子总蛋白含量、聚丙烯酰胺凝胶电泳的谱带等方面的差异。结果:蛋白质提取液提取法、TCA-丙酮法提取蛋白含量较高

但杂质多

电泳谱带严重展宽而且明显拖尾;Tris-丙酮法提出的蛋白质种类缺失严重;直接提取法提取蛋白含量少

但杂质少

电泳谱带清晰

离心2次谱带更加清晰。结论:水溶液直接提取法最适合用于聚丙烯酰胺凝胶电泳

蛋白质提取液提取法提出蛋白质含量最多。

Abstract

Objective:To determine the protein extraction method of Fructus Trichosanthis.Method:The protein yield

protein purity and polyacrylamide gel electrophoresis（PAGE） patterns were determined and analyzed.Result: More proteins were extracted by the method of protein extracted liquid and trichloroacetic acid（TCA）-ace-tone

but their bands were not clear and the resolution was low because of more impurities extracted.It waseasy to produce high-abundance proteins and interfered in the electrophoresis resolution.Protein yield and band numbers by Tris-base /acetone were the lowest.Besides

the protein content by commonly used direct extraction was low

but with less impurity

clearer bands and highest resolution.Conclusion: Direct extraction is the most suitable method for PAGE and more protein can be extracted by protein extracted liquid extraction method.
