酸枣仁合欢方中酸枣仁皂苷A的薄层色谱鉴别和含量测定
摘要
目的:建立中药复方酸枣仁合欢方中酸枣仁皂苷A的定性鉴别以及高效液相色谱-蒸发光散射检测法的含量测定方法。方法:采用薄层色谱法对方剂中的有效成分酸枣仁皂苷A进行鉴别。采用Ameritech Accurasil C18色谱柱(4.6 mm×250 mm
5μm)
流动相为甲醇-水(60∶40)
流速1.0 mL·min-1
柱温35℃
检测器漂移管温度102℃。结果:在薄层色谱图谱中可以检出酸枣仁皂苷A
斑点清晰可见
重复性好。酸枣仁皂苷A检测浓度在0.6~3.0μg与峰面积呈良好的线形关系(r=0.999 9)
平均回收率为100.16%(RSD 1.63%)。结论:定性定量方法简便、准确、重复性好
可作为酸枣仁合欢方的质量控制方法。
Abstract
Objective:To establish a TLC method for identification of Semen Zizyphi Spinosae and a HPLC-ELSD method for determination of jujuboside A in Suanzaoren Hehuan Formula.Method:Quality of Semen Zizyphi Spinosae was identified with thin layer chromatography.The analytical column was Ameritech Accurasil-ODS(4.6 mm × 250 mm
5 μm).The mobile phrase consist of methanol-water(60:40).The flow rate was 1.0 mL·min-1.The column temperature was set at 35 ℃.The tube temperature of the detector was 102 ℃.Result:TLC could be used for the identification of Semen Zizyphi Spinosae
the spots were clear and accurate.The jujuboside A was linear with-in the range of 0.6-3.0 μg(r = 0.999 9).The average recovery was 99.98% and RSD was 1.19%.Conclusion:These methods are sensitive
accurate and simple.The methods can be used for the quality control of Suanzaoren He-huan Formula.
