HPLC测定桑白皮中绿原酸的含量
摘要
目的:建立桑白皮饮片中绿原酸的含量测定方法。方法:利用星点设计法优化供试品溶液的提取条件
采用HPLC测定绿原酸的含量
Diamonsil C18柱(4.6 mm×250 mm
5μm)
流动相乙腈-0.4%磷酸(11∶89);检测波长327 nm;柱温30℃
流速1 mL·min-1。结果:供试品溶液较佳的提取条件为60%甲醇、提取时间15 min
溶媒比为10倍;绿原酸进样量范围在0.045~0.45μg
峰面积与进样量呈良好的线性关系
回归方程为Y=2 329.028 9X+8.184 1(r=0.999 9)
平均回收率为97.89%
RSD 3.14%。结论:建立的方法简便、重复性好、准确度高
可用于桑白皮饮片的质量控制。
Abstract
Objective:To establish a HPLC method for the content determination of chlorogenic acid in Mori Cortex.Method: The central composite rotatable design was used to optimize extracted conditions.The content of chlorogenic acid in Mori Cortex was determined by HPLC on a Diamonsil C18 column(4.6 mm×250 mm
5 μm)detected at the wavelength of 327 nm with acetonitrile-0.4% phosphoric acid(11∶89)solution as the mobile phase
the flow rate was 1 mL·min-1.Result: The optimum extractable conditions to obtain desirable extraction yields for chlorogenic acid were found at the methanol concentration of 60%
extraction time of 45 min and solvent quantity of 10 time
chlorogenic acid was linear in the range of 0.045-0.45 μg
regress equation was Y=2 329.028 9X+8.184 1(r=0.999 9)
the average recovery was 97.89% with RSD of 3.14%.Conclusion: The method of HPLC determination is simple
reliable and accurate and can be used for the quality control of Mori Cortex.
